In this study karyotypic features of the five species of the family Bufonidae from the central area of Argentina are described. The species are Rhinella achalensis, Rhinella arenarum, Rhinella fernandezae, Rhinella schneideri and Melanophryniscus stelzneri. The metaphases were obtained from intestinal and testis cells, using conventional techniques. Twenty metaphasic figures per individual were analyzed and the total length of each chromosome and the length of the four arms were measured. The obtained measurements were processed using Excel 2000 to obtain the average length of the arms p and q, the arm ratio, the centromeric index, the relative chromosome length and the relative arm length. All species showed karyotype 2n = 22, and karyotype formula of 6 : 5. Pairs one to six were large, with a relative chromosome length between 18.64-7.59%; pairs seven to eleven were small, with a relative chromosome length between 7.18-2.42%. In all species the chromosome morphology was metacentric or submetacentric. Karyotype and ideograms were made for all species, based on morphometric parameters of the chromosome complement. Finally, discriminant analysis was used to separate the five species analyzed, with a highly significant classification rate of 80% and P < 0.0001. These results agree, in general, with those presented by other authors, however, in M. stelzneri detailed karyological studied have not been made so far, thus this work represents a significant contribution to the karyotypic decryption features of this species and the Rhinellla species from central area of Argentina.

Fluorescence in situ hybridization (FISH) using telomeric and ribosomal sequences was performed in four species of toad genus Chaunus: C. ictericus, C. jimi, C. rubescens and C. schneideri. Analyses based on conventional, C-banding and Ag-NOR staining were also carried out. The four species present a 2n = 22 karyotype, composed by metacentric and submetacentric chromosomes, which were indistinguishable either after conventional staining or banding techniques. Constitutive heterochromatin was predominantly located at pericentromeric regions, and telomeric sequences (TTAGGG)(n) were restricted to the end of all chromosomes. Silver staining revealed Ag-NORs located at the short arm of pair 7, and heteromorphism in size of NOR signals was also observed. By contrast, FISH with ribosomal probes clearly demonstrated absence of any heteromorphism in size of rDNA sequences, suggesting that the difference observed after Ag-staining should be attributed to differences in chromosomal condensation and/or gene activity rather than to the number of ribosomal cistrons.

SummaryThe first cytogenetics of the Malayan snail-eating turtle (Malayemys macrocephala) from the Chi river basin, Khon Kaen Province, Thailand, were studied. Blood samples were taken from two male and two female turtles. Standard T-lymphocyte cell culture at 26 C for 96 h was applied. The mitotic chromosomes were harvested by colchicine-hypotonic-fixation-air drying technique. Conventional staining and GTG-banding techniques were applied to stain the chromosomes with 20% Giemsa s solution. Results showed that the number of diploid chromosomes was 2n=50, while the fundamental number (NF) was 40 in both males and females. The types of macrochromosomes were 4 metacentric, 8 submetacentric, 6 acrocentric, 4 telocentric chromosomes, and 28 microchromosomes. The GTG-banding technique showed that each chromosome pairs could be clearly differentiated and the numbers of bands in the M. macrocephala was 99. There is no observation of strangely size chromosomes related to sex. The karyotype formula is as follows:
