Genetic instability of sugarcane plants derived from meristem cultures

Abstract: The RADP (Random amplified polymorphic DNA) technique was used to detect tissue-culture-induced variations in sugarcane. Plants of the Brazilian variety RB83-5486 propagated via rhizomes and via meristem cultures were studied. The polymorphism rate for 98 RAPD loci was 6.93% when the plants derived from meristems. Besides, in order to evaluate the influence of the number of subcultures on the generation of somaclonal variation, field-grown RB83-5486 plants derived from 10 meristems were studied after five subc… Show more

“…Maritano et al (2010) suggest a distance value of 0.05 to consider that the clones obtained by in vitro culture are genetically identical, so according to the results, obtained calluses are not clones since they showed genetic variability. Zucchi et al (2002) reported polymorphism ranges of 3.06% to 5.1% between the mother plant and explants derived from sub-crops in sugarcane, polymorphism occurred in all sub-cultures and did not increase with the number thereof; as in our work, higher level of variation between the mother plant and established explants in vitro was shown. The process of in vitro culture is a stressing factor for the genome of the plant; during the cultivation in vitro, the plant suffers cell dedifferentiation, mainly in the first generation, in the next generations or sub-cultures, the level of polymorphism decreases, due to non-predictable processes (Zucchi et al 2002).…”

“…It was reported that variation may be resulted from the pre-existing variation of the explants (Wakasa 1979;Nwauzoma & Jaja 2013), the et al long period of the culture (Koornneef 1991;Masoud & Hamta 2008), the high level of plant growth regulator (Bairu 2006), the frequent et al subculture (Eeuwens 2002), the genotype et al dependence (Zucchi . 2002), activation of et al transposable elements (Bairu 2011), and et al hypo-or hypermethylation of DNA (Abdellatif et al 2012).…”
