Summary Híbrido de Timor (HT) is a natural interspecific hybrid of Coffea arabica L. with Coffea canephoraPierre ex Froehner that has played a substantial role in breeding programs as source of resistance genes. The original HT plant is represented by the anorthoploid accession CIFC 4106 , an allotriploid plant with 3x=33 chromosomes and 2C=2.10 pg. From this HT plant, other accessions have been obtained and used in crossings with C. arabica to provide resistant plants to the main pathogens. However, each HT accession and their derivates show a particular karyotype due to irregular anaphasic disjunction, which results in a few reproductive cells with an unbalanced number of chromosomes. Regarding this fact and the crop relevance of HT, this study aimed to develop a direct somatic embryogenesis system for the clonal propagation of this germplasm, using the first HT plant as explant donor. To accomplish this purpose, disinfested leaf explants of greenhouse-cultivated HT CIFC 4106 plants were inoculated in medium supplied with 0.001 g L 1 6-benzylaminopurine. Somatic embryogenesis process was asynchronous, with distinct developmental stages occurring simultaneously. Besides, emergence of secondary embryos from primary ones was observed. Mature embryos were germinated, and well-rooted embryos were selected for plantlet regeneration. The nuclear DNA content and the karyotype showed that allotriploidy was conserved in all regenerated plantlets. Considering these results, the direct somatic embryogenesis protocol adopted in the present work was imperative for the accurate clonal propagation of HT, being relevant for multiplication and conservation of elite accessions that show an irregular meiosis.

RESUMOObjetivou-se estudar o efeito da cinetina, GA 3 e ANA na indução in vitro de embriões somáticos de cafeeiro pela via direta. Segmentos foliares retirados de plântulas cultivadas in vitro foram inoculados em meio de cultura MS com 50% dos sais contendo as seguintes combinações de cinetina (0; 1; 2; 4 e 8 mg L -1 ) e GA 3 (0; 2,5; 5; 10 e 20 mg L -1 ). Os meios de cultura utilizados tiveram pH ajustado para 5,8 ± 1 antes de serem autoclavados. O experimento foi mantido em sala de crescimento a 25 ± 1°C. Avaliou-se número total de embriões somáticos, o número de embriões cotiledonares, o número de embriões torpedo e a média dos comprimentos dos embriões. A ação combinada entre cinetina, GA 3 e ANA estimulou a indução de embriões somáticos pela via direta. O maior comprimento de embriões foi observado quando se utilizou 8 mg L -1 de cinetina e 8,0 mg L -1 de ANA ou 17 mg L -1 de GA 3 e 8,0 mg L -1 de ANA isoladamente. The culture media utilized had their pH adjusted to 5,8 ± 1 before being autoclaved. The experiment was carried out growth room at 25 ± 1°C. Total number of embryos, number of embryos cotiledonar, number of embryos of torped and length of embryo were evaluated. The combination between kinetin and GA 3 promoved induction of embryos. The use of kinetin 8 mg L -1 and GA 3 17 mg L -1 not associate in medium with 8,0mg L -1 of ANA, promoter higher rates in vitro.

Somatic embryogenesis from zygotic embryos of Heloconia bihai (L.) L. cv. Lobster Claw TwoTropical flowers have contributed to the growth of the cut flower market in Brazil, especially plants of the genus Heliconia. Heliconias are usually propagated vegetatively, although this method can spread diseases and in the near future may severely comprise commercial flower production. In vitro cultivation represents an important potential alternative for guaranteeing the production of high-quality heliconia planting stocks. However, the micropropagation of these plants using stalk apices faces severe problems of endophytic contamination, making it difficult the supply of enough explants. Therefore, alternative techniques for the in vitro production of planting stock were investigated using other explant tissues to induce somatic embryogenesis. Zygotic embryos from immature and mature fruits of H. bihai (L.) L. cv. Lobster Claw Two were cultured in combinations of IAA (0; 5.70, e 11.41 µm L -1 ) and 2,4-D (0; 22.62; 45.24; 67.86 e 90.48 µm L -1 ) to induce somatic embryos during 90 days of culture. Histological analyses showed that somatic embryos were formed only with zygotic embryos derived from mature fruits cultivated in the absence of growth regulators.
