Introdução: A resistência de espécies fúngicas às drogas usualmente empregadas no meio clínico é motivo de grande interesse na área médica. Objetivo: Avaliar susceptibilidade e resposta in vitro de espécies de Trichophyton spp. a drogas antifúngicas de interesse em clínica médica. Métodos: Foram utilizadas 12 amostras de isolados clínicos de humanos, sendo nove de T. mentagrophytes e três de T. tonsurans. Foram realizados testes de susceptibilidade segundo os métodos de difusão em ágar (DA) e microdiluição em caldo (MC). Resultados: No método de DA, a espécie T. tonsurans apresentou percentual de sensibilidade de 33% em relação à anfotericina B e de 66% ao itraconazol, com 100% de resistência frente ao cetoconazol e ao fluconazol. A espécie T. mentagrophytes também apresentou 100% de resistência frente ao cetoconazol nesta técnica, com 11% de sensibilidade ao cetoconazol, 22% ao itraconazol e 22% das amostras classificadas como sensível dose dependente. No método de MC, a espécie T. tonsurans apresentou percentual de sensibilidade de 66%, 55% e 33% em relação ao cetoconazol, fluconazol e itraconazol, respectivamente. A espécie T. mentagrophytes apresentou percentuais de sensibilidade de 11%, 11%, 33% e 55% para anfotericina B, itraconazol, cetoconazol e fluconazol, respectivamente. Conclusão: Houve resistência in vitro das espécies do T. mentagrophytes e T. tonsurans frente ao antifúngico fluconazol e resistência relativa frente ao cetoconazol no método de DA. Na MC, no entanto, foram observados importantes percentuais de sensibilidade das duas espécies analisadas frente aos antifúngicos fluconazol e cetoconazol quando comparadas ao itraconazol e à anfotericina B.

Dermatophytoses are mycoses that affect keratinized tissues in both humans and animals. The aim of this study was to investigate the antifungal activity of the oleoresin extracted from Copaifera langsdorffii Desf. against the strains Microsporum canis ATCC 32903, Microsporum gypseum ATCC 14683, Trichophyton mentagrophytes ATCC 11481 and Trichophyton rubrum CCT 5507. The antimicrobial activity was determined by minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) values. Ketoconazole and terbinafine were used as reference drugs. The copaiba oleoresin showed moderate fungicidal activity against T. mentagrophytes ATCC 11481 (MIC and MFC = 170 μg mL ). There was no activity against M. canis ATCC 32903 and M. gypseum ATCC 14683. SEM analysis revealed physical damage and morphological alterations such as compression and hyphae clustering in the structure of the fungi exposed to the action of the oleoresin. The results stimulate the achievement of in vivo assays to confirm the benefits of the application of oleoresin extracted from copaiba in the treatment of dermatophytosis, both in humans and in animals.

The stability of pharmaceuticals is influenced by several factors, directly related to the product, or to environmental factors, such as the presence of light and oxygen. Ketoconazole is an antifungal drug of the azole class that is prone to degradation processes, such as oxidation and hydrolysis, changing from white to pink, suggesting the formation of degradation products. Capuchin (Tropaeolum majus L.) is a species of pharmacological interest due to its phytochemical composition; it is rich in vitamin C and flavonoids with known antioxidant activity. The objective of this work was to investigate the antioxidant activity of capuchin hydroethanolic extracts and evaluate their influence on the stability of ketoconazole. For the evaluation of the antioxidant activity of the extracts, the DPPH radical was used. Different ketoconazole gel formulations were manipulated and subjected to a stability study to compare the antioxidant efficiency of the extract and sodium metabisulphite by analyzing the samples by ultraviolet scanning. Microbiological assays were performed to evaluate the activity of ketoconazole against the fungus Candida albicans. The extract solution presented antioxidant activity equivalent to a 12.71 μg/mL metabisulfite solution, and a 0.085 μg metabisulfite/μg extract ratio was calculated. In the spectrophotometric analysis, it was observed that the absorption spectrum of the extractcontaining formulation presented differently from the others, with a possible advantage in relation to the synthetic antioxidant. In the microbiological analysis, only the samples that remained at room temperature formed an inhibition halo. Thus, the potential of a natural active was evaluated, representing a possible alternative for application in pharmaceutical formulations.
