Ehrlichiosis is a worldwide distributed disease caused by different bacteria of the Ehrlichia genus that are transmitted by arthropod vectors. Its occurrence in dogs is considered endemic in several regions of Brazil. Regarding cats, however, few studies have been done and, consequently, there is not enough data available. In order to detect Ehrlichia spp. in cats from the central-western region of Brazil, blood and serum samples were collected from a regional population of 212 individuals originated from the cities of Cuiabá and Várzea Grande. These animals were tested by the Immunofluorescence Assay (IFA) and the Polymerase Chain Reaction (PCR) designed to amplify a 409 bp fragment of the dsb gene. The results obtained show that 88 (41.5%) cats were seropositive by IFA and 20 (9.4%) cats were positive by PCR. The partial DNA sequence obtained from PCR products yielded twenty samples that were found to match perfectly the Ehrlichia canis sequences deposited on GenBank. The natural transmission of Ehrlichia in cats has not been fully established. Furthermore, tick infestation was not observed in the evaluated cats and was not observed any association between age, gender and positivity of cats in both tests. The present study reports the first serological and molecular detection of E. canis in domestic cats located in the endemic area previously mentioned.

ResumoO presente estudo avaliou a ocorrência da Erliquiose Monocítica Canina e Anaplasmose Trombocitotrópica Canina em 77 cães atendidos no Hospital Veterinário (HOVET) da Universidade Federal de Mato Grosso (UFMT) no ano de 2009, buscando associar à possíveis dados clínicos e hematológicos. Os cães foram avaliados pela amplificação parcial do genes dsb e 16S rRNA de Ehrlichia canis e Anaplasma platys e Reação de Imunofluorescência Indireta (RIFI) para E. canis. DNA de E. canis foi detectado em 18 (23,3%) e de A. platys em 07 (9,1%) cães. Cinquenta e quatro (70,1%) cães foram soropositivos, com títulos variando de 40 a 327.680. Observou-se maior frequência de positivos pela PCR para E. canis nos cães com idade até 12 meses, com anemia, linfopenia e trombocitopenia (P≤ 0,05). Dentre os positivos para A. platys, os cães leucopênicos apresentaram tendência à positividade pela PCR (P=0,07). Segundo os resultados da RIFI, cães apresentando trombocitopenia e hiperproteinemia foram associados a soropositividade (P≤0,05). Dentre os animais positivos pela PCR para E. canis, a linfoadenopatia e alterações pulmonares foram observada em 15 (30,6%) e 4 (57,1%) cães respectivamente (P≤0,05). Outras alterações não apresentaram associação significativa (P>0,05) para E. canis e A. platys. E. canis foi a única espécie de Ehrlichia detectada em cães atendidos no HOVET no ano de 2009, apresentando alta taxa de infecção em cães jovens e estatisticamente associada a cães anêmicos e trombocitopênicos. Por outro lado, A. platys apresentou baixa ocorrência entre os cães analisados. In this chapter, we describe that naturally infected dogs with Anaplasmataceae show altered rhreological parameters. Also, we have showed that lower viscosity correlated with the lower erythrocyte number and release of IFN-γ. The rheometry of the fresh blood samples was measured by using the Modular Compact Rheometer-MCR 102 (Anton Paar® GmbH, Ostfildern, Germany), and the graphs were obtained using Rheoplus software. Blood count data were obtained by analysis in a private laboratory. Diagnostic confirmation was obtained by molecular PCR technique that was used to determine the groups of not infected and infected by Anaplasmataceae. Serum cytokines were dosed by flow cytometry (FACScalibur BD®) using BD® Biosciences Cytometric Bead Array (CBA) Human Th1/Th2/Th17 Cytokine kits. The results showed a correlation between blood viscosity (p < 0.05, r = 0.73) and shear rate (p < 0.05; r = −0.676) with IFN-γ in the group of infected dogs that presented anemia, as well as correlations of shear rate with erythrocytes (p < 0.05; r = −0.88). Thus, IFN-γ appears to play an important role in the immunomodulation of the rheological behavior of naturally infected dogs to Anaplasmataceae. The alterations in cytokines profile and their relationship with blood viscosity and hematological parameters was related in this study the first time of dogs naturally infected with Anaplasmataceae.
