“…Overall donkeys tend to have more morphologically normal sperm than stallions, and a review of various studies using different donkey breeds showed that most donkeys have ≤15% morphologic defects 58 . Sperm morphology can be performed with a wet-mount preparation with semen fixed in temperature matched buffered formalin, or dry-mount preparation where smears are stained with eosin-nigrosin, Karras, or Romanoswsky 58,[60][61][62] . Additionally, a new system (Trumoph Proiser, Valencia, Spain) was recently introduced to evaluate sperm morphology.…”

“…Sperm morphology has rarely been linked with infertility in donkeys 58 , and one infertile mix-breed donkey was described to have olizoospermia, and asthenozoospermia with most sperm having proximal droplets had microtubules in disarray 60 . Currently, the authors' preference is to describe the sperm morphological defects according to the type and region of the sperm using wetmount preparations and a differential interference contrast microscope; however, there are many ways to classify sperm morphologic abnormalities (e.g., primary and secondary defects, major and minor defects, compensatory and non-compensatory) 58,61 .…”

“…Conventionally, the sperm motility and morphologic analysis are used as quick tests for semen samples. However, they do not show alterations when the morphological defects are expressed in a nanometer scale (Crespilho et al 2006), which are revealed by electron microscopy. Even with the advances of this technique, there is little data regarding the damage induced by the cryopreservation in sperm ultrastructure, and most of them are directed to domestic animals (Silva et al 2009).…”
