Oxymatrine ameliorates renal ischemia-reperfusion injury from oxidative stress through Nrf2/HO-1 pathway

Abstract: PURPOSE:To investigate if oxymatrine pretreatment could ameliorate renal I/R injury induced in rats and explore the possible role of oxymatrine in Nrf2/HO-1 pathway. METHODS:Unilaterally nephrectomized rats were insulted by I/R in their left kidney. Twenty four rats were randomly divided into three groups: sham group, I/R + saline-treated group, I/R + OMT-treated group. Oxymatrine or vehicle solution was administered intraperitoneally injected 60 min before renal ischemia, respectively. Renal function, histolo… Show more

“…Previous studies have demonstrated that, in sustained diabetes, states of hyperglycemia and oxidative stress in vivo, and non-enzymatic glycation (glycosylation) reactions are evident, which are important in the pathogenesis of diabetes and nephropathy (3)(4)(5)(6). Jin et al reported that C-type natriuretic peptide ameliorates IRI-induced acute kidney injury through the inhibition of oxidative stress (7).…”

“…HO-l is a microsomal oxidase with various functions in the process of heme metabolism (10). HO-1 is a myocardial protection factor and has anti-inflammatory, anti-oxidant, anti-apoptotic and anti-arrhythmia properties.…”

“…When cells are affected by oxidative stress or electrophilic compound stimulation, Nrf2 will translocate into the nucleus and bind to an antioxidant response element to induce the expression of genes required for the antioxidant defense system (9). The antioxidant defense system is the main protective mechanism against oxidative damage, resulting in the neutralization of oxidants and electrophiles by antioxidants in the cells (10).…”

“…Therefore, oxidative stress can cause disruptions in the normal mechanisms involved in cellular signaling pathways [1]. The nuclear factor erythroid 2-related factor 2 (Nrf2)/antioxidant response element (ARE) pathway has been found to be correlated to antioxidative stress and the scavenging of reactive oxygen species (ROS) under the condition of oxidative stress [2].…”
