Heart and systemic effects of statin pretreatment in a rat model of abdominal sepsis. Assessment by Tc99m-sestamibi biodistribition

Abstract: PURPOSE:To evaluate the heart and the Tc-99m-sestamibi biodistribution after statin pretreatment in a rat model of abdominal sepsis. METHODS:Twenty-four Wistar rats were randomly distributed into four groups (n=6 per group): 1) sepsis with simvastatin treatment, 2) sepsis with vehicle, 3) sham control with simvastatin and 4) sham control with vehicle. 24 hours after cecal ligation and puncture rats received 1.0MBq of Tc-99m-sestamibi i.v. 30min after, animals were euthanized for ex-vivo tissue counting and myo… Show more

“… 28 , 29 , 30 , 31 , 32 Statin treatment also altered biodistribution of Tc-99m-sestamibi in rats with abdominal sepsis, interpreted as a protective effect because of statin-mediated increased tissue perfusion. 33 The protective effect of statins is attributed to their effects on insulin sensitivity, improving insulin signaling in peripheral tissues, 28 preserving cardiac function and hemodynamic status, 29 , 31 reversing inflammatory cytokines levels, 15 , 16 , 17 and enhancing bacterial clearance. 17 Additionally, statins appear to decrease nitric oxide production and restore impaired vascular responsiveness in sepsis.…”

“…In the present study, we compared the effects 0.9% saline, modified coconut water (Na 3%) and water coconut 3% + Na + simvastatin, and normal coconut water on serum cytokines, biochemical and white blood cell count in rodent model with hemorrhagic shock and abdominal sepsis. Experimental models of sepsis and hemorrhagic shock has often been employed in other estudies [30][31][32] . In the present study, we found that treatment of rats with sepsis and shock with coconut water modified with sodium 3% + simvastatin reduced serum cytokines, biochemical parameters and peripheral leukocytes.…”

“…The rats (n=4) were anesthetized with xylazine associated with ketamine at doses of 10 and 70 mg/kg, respectively, intramuscular. The induction of sepsis by cecal ligation and puncture (CLP) followed the technique used in other studies [16][17][18][19] . Briefly, after shaving and disinfecting the lower quadrants of the abdomen with…”
