Radix Polygalae is a traditional Chinese medicine that has been used as a sedative and to improve memory for a number of years. The impact of Radix Polygalae in patients with Alzheimer's disease has been investigated. However the mechanisms underlying its effects remain unclear. In the current study, the toxicity of various doses (100, 40, 20, 10, 5 and 0 µg/ml) of Radix Polygalae was measured in the human neuroblastoma cell line (SH-SY5Y) using an MTT assay. Changes in amyloid β (Aβ) levels in the supernatant of Chinese hamster ovary (CHO) cells overexpressing β-amyloid pro-protein (APP) and BACE1 (CHO-APP/BACE1), were detected using an ELISA assay. In order to confirm that the Aβ reduction was associated with autophagy, the autophagy marker protein, light chain 3 (LC3), was measured by western blot analysis and autophagosomes were assessed using MDC staining. In addition, the mechanism underlying the autophagy induced by Radix Polygalae was analyzed using western blotting to measure the protein expression of mammalian target of rapamycin (mTOR), p70s6k, Raptor, protein kinase B and adenosine monophosphate-activated protein kinase (AMPK), in addition to the phosphorylated forms of these proteins. The results demonstrated no significant toxicity of Radix Polygalae in SH-SY5Y cells, at a dose of 100 µg/ml. The secretion of Aβ was markedly reduced following treatment with Radix Polygalae, and this reduction occurred in a dose-dependent manner. The autophagy levels were shown to be enhanced in the drug treatment group, using fluorescence microscopy. In addition, levels of LC3Ⅱ/LC3Ⅰ, the marker protein of autophagy, were also increased. The results of the current study suggest that Radix Polygalae may aid in the elimination of the Aβ peptide, via the induction of autophagy, by the AMPK/mTOR signaling pathway. These results may provide a basis for further kin vivo investigation.

Widespread antibiotic resistance is on the rise and current therapies are becoming increasingly limited in both scope and efficacy. Methicillin-resistant Staphylococcus aureus (MRSA) represents a major contributor to this trend. Quorum sensing controlled virulence factors include secreted toxins responsible for extensive damage to host tissues and evasion of the immune system response; they are major contributors to morbidity and mortality. Investigation of botanical folk medicines for wounds and infections led us to study Schinus terebinthifolia (Brazilian Peppertree) as a potential source of virulence inhibitors. Here, we report the inhibitory activity of a flavone rich extract “430D-F5” against all S. aureus accessory gene regulator (agr) alleles in the absence of growth inhibition. Evidence for this activity is supported by its agr-quenching activity (IC50 2–32 μg mL−1) in transcriptional reporters, direct protein outputs (α-hemolysin and δ-toxin), and an in vivo skin challenge model. Importantly, 430D-F5 was well tolerated by human keratinocytes in cell culture and mouse skin in vivo; it also demonstrated significant reduction in dermonecrosis following skin challenge with a virulent strain of MRSA. This study provides an explanation for the anti-infective activity of peppertree remedies and yields insight into the potential utility of non-biocide virulence inhibitors in treating skin infections.

RESUMO:A Schinus terebinthifolius Raddi popularmente conhecida como "aroeira" é amplamente utilizada na medicina popular, sendo seu uso relacionado, por exemplo, aos variados polifenóis distribuídos desigualmente em suas estruturas, como folhas, cascas, frutos, flores e sementes. Esta planta atua no tratamento de afecções como diarreias, gastrites e dispepsias; tem ação adstringente, tônico, estimulante e anti-neurágico. Utilizada por via oral ou tópica para cura de várias doenças, com efeitos diversos, incluindo ações antimicrobianas, antiinflamatórias e cicatrizantes. Experimentos realizados com a espécie em questão são facilitados, pois além de apresentar um uso amplo na medicina popular brasileira, ela possui fácil extração, secagem, moagem, preparo, conservação e aplicação. A S. terebinthifolius Raddi apresenta-se como uma opção de baixo custo, acessível, todavia sua manipulação se faz muitas vezes sem embasamento científico. Este artigo faz um levantamento amplo dessa planta mostrando seus efeitos benéficos relacionados à medicina popular. Palavras-chave: Conhecimento popular; Medicina comunitária; Vegetal; Fitoterápico ABSTRACT:The Schinus terebinthifolius Raddi popularly known as "Aroeira" is widely used in folk medicine, being your use related, for example, the various polyphenols unevenly distributed in their structures, such as leaves, bark, fruits, flowers and seeds. This plant operates in the treatment of diseases such as diarrhea, gastritis and dyspepsia; action is astringent, tonic, stimulant and anti-neuralgic. Used orally or topically for curing various diseases, including various effects including antimicrobial, anti-inflammatory and healing actions. Experiments with the species in question are facilitated, as well as presenting a wide use in Brazilian folk medicine, it has easy extraction, drying, grinding, preparation, storage and application. The S. terebinthifolius Raddi presents itself as a low cost option available, but its handling is done often without scientific basis. This article makes a comprehensive study of this plant showing its beneficial effects related to folk medicine.
