Simvastatin attenuates neutrophil recruitment in one-lung ventilation model in rats

Abstract: PURPOSE:To investigate the anti-inflammatory effects of simvastatin in rats undergoing one-lung ventilation (OLV) followed by lung re-expansion. METHODS:Male Wistar rats (n=30) were submitted to 1-h OLV followed by 1-h lung re-expansion. Treated group received simvastatin (40 mg/kg for 21 days) previous to OLV protocol. Control group received no treatment or surgical/ventilation interventions.Measurements of pulmonary myeloperoxidase (MPO) activity, pulmonary protein extravasation, and serum levels of cytokine… Show more

“…Simvastatin is a cholesterol-lowering agent that reduces local and systemic inflammatory responses through unknown mechanisms 6 Previous data has suggested that simvastatin blocks the activities of many important proinflammatory cytokines, 7,8 such as TNF-alpha and NF-κB. These cytokines are found to be actively involved in apoptosis.…”

“…Previous studies have found that simvastatin inhibited the activities of numerous important pro-inflammatory factors, including TNF-α and NF-κB ( 16 , 23 ). TNF-α can activate the NF-κB signaling pathway, which is involved in apoptosis ( 24 ).…”

“…Furthermore, simvastatin is effective in the treatment of inflammation by directly or indirectly triggering pro-inflammatory signaling pathways ( 15 ). Numerous studies have reported that simvastatin prevented fibrosis in certain types of injured tissues, including articular cartilage, lungs, kidneys, heart and blood vessels ( 16 ). In addition, certain studies have demonstrated that simvastatin enhanced the survival of RGCs by regulating the anti-apoptotic protein B-cell lymphoma 2 (Bcl-2) in a model of IR ( 17 , 18 ).…”
