The effect of 6% Hydroxyethyl starch vs. Ringer's lactate on acute kidney injury after renal ischemia in rats

Abstract: PURPOSE:To compare fluid replacement therapy with Hydroxyethyl starch 6% (HES) versus Ringer's lactate (RL) in a rodent model of non-septic renal ischemia. METHODS:Forty male Wistar rats were randomized to receive HES 2 ml.kg -1 .hr -1 or RL 5 ml. kg -1 .hr -1 that underwent 30 minutes of renal ischemia followed by reperfusion. Twelve hours after kidney ischemia, the kidneys were evaluated for histological changes. Serum NGAL levels were obtained at different times of the experimental protocol. RESULTS:Rodents… Show more

“…This study is an experimental model for renal I/R investigation and has limitations. A single kidney model was utilized, 15,16 since there were right nephrectomies to prevent hyper activation of the renin-angiotensin-aldosterone system and severe vasoconstriction on the contralateral kidney. 24 There are still uncertainty about the best methods for testing and several drugs under investigation to prevent I/R lesions, like Allopurinol, 22 hence, some long way before appropriate human studies.…”

“…The preparation was based on a previous described method for renal I/R. 15,16 Animals were cared for according to the guidelines of the Council for International Organization of Medical Sciences (CIDMS) ethical code for animal experimentations. Animals fasted overnight prior to the experiment but were given free access to water.…”

“…Histological evaluation was performed under light microscopy (Zeiss Axioplan 2 Microscope; Zeiss, Germany) by two study-blinded pathologists. Renal damage was assessed for glomerular, tubular, interstitial and vascular lesions, and papillary necrosis following the classification previously used by other researchers [ 9 11 34 ]. A grade from 0 to 3 was attributed for the majority of the renal lesions: Grade 0 (absence of lesion), Grade 1 (< 25% of the kidney area affected), Grade 2 (25–50% of the kidney area affected), and Grade 3 (> 50% of the kidney area affected).…”
