Abstract: Purpose: To investigate blood creatinine and renal histology in rats anesthetized with S(+)-ketamine (keta) or dexmedetomidine (dex) and submitted to kidney ischemia/reperfusion injury (IRI). Methods: Under intraperitoneal (ip) S(+)-ketamine, 20 maleWistar rats were divided into two groups (n=10): maintenance with iv S(+)-ketamine or dex (keta and dex groups), and submitted to right (R) nephrectomy and left (L) renal artery clamping for 45 min. Blood creatinine was measured before ischemia (T1) and 48h after r… Show more

“…α 2B -AR stimulates protein kinase C (PKC) activity and inositol triphosphate production in renal distal convoluted tubule cells. This demonstrates that PKC is important for ischemic preconditioning since it activates the intracellular signaling pathways providing organ protection, the opening of sarcolemmal and mitochondrial ATP-sensitive K + channels and through gene transcription, promotes the protective cellular protein synthesis (21). DEX reverses the suppression of HSP27 phosphorylation by the activation of the adenylyl cyclase-cyclic adenosine monophosphate (cAMP) system in C6 cells, suggesting that DEX may have a neuroprotective effect through the modification of PKC activation-induced HSP27 phosphorylation (22).…”

“…Dexmedetomidine (Dex) is a potent agonist for all three subtypes of human α 2 -ARs, having sedative, analgesic, and anti-sympathetic effects [11]. Dex has been shown to provide good perioperative hemodynamic stability and to have a protective effect on specific organs, including the heart, brain, and kidney, probably via decreasing cell apoptosis in a Bax/Bcl-2-related manner [13] and inhibiting pro-inflammatory cytokine release [1416]. …”

“…10 Another one, also employing an I/R model in rats, found a correlation between the use of dexmedetomidine and a reduction of serum urea and creatinine levels, 11 whereas in other reduced serum creatinine levels was found, without protection from injury. 12 The aim of the present study was to assess the effect of dexmedetomidine in an experimental I/R model in rats through histological examination and measurement of the serum levels of neutrophil gelatinase-associated lipocalin (NGAL), as an early biomarker for AKI. 13,14…”
