Mast cells and angiogenesis in canine mammary tumor

Abstract: The correlation between microvessel density and mast cells density in canine mammary tumors was studied. Sixty-five samples of canine mammary tumors, being 24 benign and 41 malignant, were analyzed. The routine Toluidine Blue staining method was used to assess the mast cells. To evaluate angiogenesis, the immunohistochemical expression of CD31 was assessed. There was no significant difference in either mast cells (P=0.44) or microvessel density (P=0.77) between malignant and benign tumors. A positive correlati… Show more

“…The blocked sections were incubated overnight at 48°C anti-mouse primary antibodies for TP PD-ECGF AB1 (clone PGF 44.C) (Neo Markes, Freemount, California, USA, diluted 1/100) (15) and monoclonal mouse antibody anti-CD31 Clone JC70A (Dako, Glostrup, Denmark) diluted 1/50, for 40 min then thoroughly washed in 0.05 M buffered with Tris saline (pH = 7.6) and incubated with streptavidin-peroxidase (Dako, 1: 100) for 40 min. 3, 3- diaminobenzidine (DAB) (Dako Glostrup, Denmark) was used as the chromogen; to counteract the core of Gill'S hematoxylin (Polysciences, Warrington, PA, USA) after sections have been dehydrated and assembled (34, 35). We do also have evidence of negative controls not immunostained and positive control of not tumor cells from canine expressing TP that show sporadic area of immunostaining.…”

“…Moreover, IL-28 secreted by the MDSCs stimulates the STAT3 pathway which increases VEGF-C expression and therefore induces angiogenesis [55]. A positive correlation between microvessel density and mast cells in malignant canine mammary cancer was found, which, in turn, suggests that these immune cells play an important role in canine mammary cancer angiogenesis, similar to human breast cancer [53]. TAMs in canine mammary cancer have already been associated, by our group, with skin ulceration, histological type, nuclear grade, tubular differentiation, and decrease in overall survival [56].…”

“…The presence and activity of mast cells in the tumor microenvironment remain controversial. Some studies reported an association between high number of profile cells of mast cells in the tumor microenvironment, greater release of angiogenic factors and high number of profile cells of microvasculature in human and canine mammary carcinomas (Cimpean et al, 2017;Lavalle et al, 2010). On the other hand, Amini et al (2007), Dabiri et al (2004) and Rajput et al (2008) observed, in human mammary carcinomas, that a high number of profile cells of mast cells was related to low grade tumors and estrogen receptor positivity, to patients without lymph node involvement and to patients with better prognosis, respectively.…”
