The aim of this study was to examine differences in the microbiological profile and antimicrobial resistance of bacteria isolated from milk from organic and conventional sheep and goat farms. Twenty-five organic and 25 conventional sheep and goat farms in the region of Thessaly, Greece participated in this study. A standardised detailed questionnaire was used to describe farming practices. A total of 50 samples were collected and analysed for total viable count (TVC), total coliform count (TCC) and somatic cell count (SCC), while Staphylococcus aureus and Escherichia coli were isolated using standard methods. Isolates were identified at species level by Api-test and Matrix-Assisted Laser Desorption/Ionisation-Time of Flight Mass Spectrometry (MALDI-TOF MS). Susceptibility to a panel of 20 for E. coli and 16 for S. aureus antimicrobials was determined by the agar dilution method. Pulsed Field Gel Electrophoresis (PFGE) was performed for S. aureus and E. coli isolates to determine predominant clones. Lower counts of TVC, TCC and SCC were identified in milk from the organic farms, possibly due to differences in the hygienic farming practices found on those farms. API-tests and MALDI-TOF MS showed no significant differences in the S. aureus and E. coli isolates. Overall, antimicrobial resistance rates were low, while a statistically higher percentage was estimated among strains originating from conventional farms in comparison with organic farms, possibly due to the restriction of antibiotic use in organic farming. PFGE revealed diversity among S. aureus and E. coli populations in both organic and conventional farms indicating circulation of 2-3 main clones changing slightly during their evolution. Consequently, there is evidence that milk from the organic farms presents a better microbiological profile when compared with milk from conventional farms.

Antibiotic resistance has increased in recent years, raising the concern of public health authorities. We conducted a study of Escherichia coli isolates obtained from human and food samples to assess the prevalence of antimicrobial resistance and to determine the genotype and clonal relationship of 84 E. coli isolates (48 from humans and 36 from foods). An antimicrobial susceptibility test was performed using the disk diffusion method. Virulence factors were evaluated by multiplex PCR, and the clonal relationship among the resistant isolates was studied by Pulsed Field Gel Electrophoresis (PFGE). All isolates were susceptible to ceftriaxone. Overall, 26%, 20.2%, 15.4% and 6% of the isolates were resistant to tetracycline, ampicillin, sulfamethoxazole/trimethoprim and cephalotin, respectively. Twenty two percent of the isolates exhibited resistance to more than one antimicrobial agent. Multiple-drug resistance was mostly observed in the human isolates and involved the antibiotics ampicillin and tetracycline. None of the six virulence genes were identified among the isolates. Analysis of genetic diversity by PFGE of 31 resistant isolates, revealed 29 distinct restriction patterns. In conclusion, E. coli from humans and foods are resistant to commonly used antibiotics and are highly genetically diverse. In this setting, inappropriate use of antibiotics may be a cause of high resistance rate instead of clonal spread.

RESUMOUm total de cinquenta e nove queijos muçarela de fabricação caseira foi coletado em um pequeno produtor no vale do Jequitinhonha (Nordeste de Minas Gerais, Brasil) e 147 isolados de E. coli recuperados dos queijos foram examinados para sua resistência frente a 12 agentes antimicrobianos. As principais resistências foram detectadas para tetraciclina (52,4%), cefalotina (44,2%), amicacina (34,1%), ampicilina (31,3%) e ácido nalidíxico (30,0%). Porcentagem elevada de resistência a múltiplas drogas (60,0%) foi encontrada, o que representa um motivo de preocupação devido ao risco de disseminação de genes de resistência aos antimicrobianos na microbiota humana. SUMMARYA total of 59 home-made mozzarella cheese samples were taken at a local producer in the Jequitinhonha Valley (Northeast of Minas Gerais, Brazil). A hundred-forty-seven E .coli isolates from the cheese were screened for resistance to 12 antimicrobial agents. Predominant resistance was to tetracycline (52.4%), cefalothin (44.2%), amikacin (34.1%), ampicillin (31.3%) and nalidixic acid (30.0%). Multidrug resistance was found in high level (60.0%) which represents a reason for concern due to the risk of dissemination of antimicrobial resistant genes to the microbiota of human beings.
