Synergistic antitumor effect of TRAIL and adriamycin on the human breast cancer cell line MCF-7

Abstract: The aim of the present study was to determine the effect of the combination of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) and adriamycin (ADM) on the human breast cancer cell line MCF-7 and to identify potential mechanisms of apoptosis. Cell viability was analyzed by the MTT assay and the synergistic effect was assessed by the Webb coefficient. Apoptosis was quantified using the annexin V-FITC and propidium iodide staining flow cytometry. The mRNA expression of TRAIL receptors was measured… Show more

“…Thus, it is important to develop new strategies to overcome this type of resistance in tumor cells. Interestingly, some groups have described the ability of subtoxic concentrations of chemotherapeutic drugs to sensitize tumor cells resistant to TRAIL [23,25,26]. Also the anticancer efficacy of TRAIL against BC cells is known to be retained in the bone microenvironment, even in the present of biologically active OPG at a supraphysiologic concentration [18].…”

“…TRAIL, which is able to selectively induce apoptosis in cancer cells, is a potential targeted drug for cancer therapy (20). However, its clinical use is limited by cellular resistance to cell death which occurs in ~50% of cancer cells (21).…”

“…The sensitivity of cells to drug is evaluated by the value of IC 50 . IC 50 < 10  μ g/mL, which indicates that cells are sensitive to drug, while IC 50 ≥ 10  μ g/mL, which suggests that cells are relatively resistant to drug [20]. …”
