Effects of 60Co gamma radiation on crotamine

Abstract: Ionizing radiation can change the molecular structure and affect the biological properties of biomolecules. This has been employed to attenuate animal toxins. Crotamine is a strongly basic polypeptide (pI 10.3) from Crotalus durissus terrificus venom composed of 42 amino acid residues. It induces skeletal muscle spasms leading to a spastic paralysis of hind limbs in mice. The objective of the present study was to carry out a biochemical study and a toxic activity assay on native and irradiated crotamine. Crota… Show more

“…This agrees with the findings of other authors (7,12,21,(31)(32)(33)(34)(35)(36)(37)(38)42), which assured that ionizing radiation has the property to attenuate venoms toxicity without altering immunological properties.…”

“…The principal physical agents are: x-ray radiation (19), ultraviolet radiation (46), heat (15), and gamma radiation (25). The latter, according to previous works (1,7,9,12,14,17,21,(31)(32)(33)(34)(35)(36)(37)(38)(41)(42)(43), has proved to be an excellent tool of detoxification, since it is able to reduce toxicity without altering immunogenicity. In addition, this radiation does not add any new substance to the venom.…”

“…Indeed, the lethality of unfolded crotamine was shown to be reduced by approximately 50%. 12 Interestingly, crotamine appears to be active against only a narrow spectrum of bacteria, in contrast with most vertebrate b-defensins that are broadly active. This is perhaps not surprising, given a great degree of diversity in the primary amino-acid sequence between crotamine and b-defensins (Figure 1).…”

“…The crotalic venom from the snake Crotalus durissus terrrificus was purchased from CEVAP, Botucatu, São Paulo, Brazil, and used to purify crotamine as described, 12 with slight modifications. Briefly, crotamine was purified from crude venom by size exclusion on a Sephacryl S200 column (GE Healthcare, Uppsala, Sweden), followed by cation exchange chromatography on a 1-ml Resource S on FPLC system (Akta Purifier System, GE Healthcare).…”

“…The activation of snake venoms occurs under the influence of some physical factors, for example, under the influence of ultraviolet irradiation and X-rays [12]. So, in 7 days after a venom irradiation of Filipina cobras by γ radiation ( 60 Со) in doses 0.25, 0.5, 1 mrad, toxicity of snake venom made 83, 66 and 43 %, accordingly from not irradiated venom [16,17,18]. On literary data snake venom in the dried up kind keeps pharmacological and toxic properties till 22 years and even more.…”
