Angiotensin II-mediated vascular smooth muscle cell growth signaling

Abstract: The mechanism by which Ang II stimulates the growth of vascular smooth muscle cells was investigated by measuring the phosphorylation of mitogen-activated protein kinases ERK 1 and ERK 2. Ca 2+ ionophore was found to have effects practically analogous to Ang II. We found that the signaling pathway involves the activation of epidermal growth factor receptor (EGFR) kinase, activation of the adaptor proteins Shc and Grb2, and the small G-protein Ras. Although the mechanism of AT 1 -(or Ca 2+ )-induced activation … Show more

“…Increases in pERK1/2 confirmed that WNK3 knockout mice do not lack the AngII response in aorta ( Figure S2). 25 These data indicate Figure 2. Phosphorylation of STE20/SPS1-related proline/ alanine-rich kinase (SPAK) and Na-K-Cl cotransporter isoform 1 (NKCC1) in aorta from wild-type (WT) and with-no-lysine kinase 3 (WNK3) knockout (KO) mice fed a normal diet or a low-salt diet.…”

“…GPCR -epidermal growth factor receptor (EGFR) cross-talk pathways are widely established and it has been reported that stimulation of angiotensin II involves transactivation of the EGFR, resulting in the activation of the MAPK pathway in various cell types (16,(41)(42)(43)(44)(45).…”

“…It has been recently suggested that Ras induces premature senescence in vascular cells, thereby promoting human atherosclerosis [23]. It is well recognized that Ang II mediates cell growth via a Ras-dependent mechanism [36]. It has also been reported that Aldo rapidly induces Ras protein level through a genomic mechanism, thereby promoting cell proliferation [5].…”
