Austropuccinia psidii, the causal agent of myrtle rust, is a biotrophic pathogen whose growth and development depends on the host tissues. The uredospores of A. psidii infect Eucalyptus by engaging in close contact with the host surface and interacting with the leaf cuticle that provides important chemical and physical signals to trigger the infection process. In this study, the cuticular waxes of Eucalyptus spp. were analyzed to determine their composition or structure and correlation with susceptibility/resistance to A. psidii. Twenty-one Eucalyptus spp. in the field were classified as resistant or susceptible. The resistance/susceptibility level of six Eucalyptus spp. were validated in controlled conditions using qPCR, revealing that the pathogen can germinate on the eucalyptus surface of some species without multiplying in the host. CG-TOF-MS analysis detected 26 compounds in the Eucalyptus spp. cuticle and led to the discovery of the role of hexadecanoic acid in the susceptibility of Eucalyptus grandis and Eucalyptus phaeotricha to A. psidii. We characterized the epicuticular wax morphology of the six previously selected Eucalyptus spp. using scanning electron microscopy and observed different behavior in A. psidii germination during host infection. It was found a correlation of epicuticular morphology on the resistance to A. psidii. However, in this study, we provide the first report of considerable interspecific variation in Eucalyptus spp. on the susceptibility to A. psidii and its correlation with cuticular waxes chemical compounds that seem to play a synergistic role as a preformed defense mechanism.

When investigating the potential use of plants as a raw material for an all-natural cosmetic formulation, the main parameters are the chemical composition, antioxidant potential, antimicrobial action, and toxicity. Additionally, the production of natural cosmetics should also consider the availability of primary materials and the environmental and socioeconomic impact. Gliricidia sepium is a species that produces a large amount of plant material, being cultivated in the agroforestry system. However, studies of phytochemical composition and chemical spatial distribution are scarcely using the MALDI MS (matrix-assisted laser desorption ionization mass spectrometry) and MALDI MSI (mass spectrometry imaging) techniques. A methodology was developed to optimize ionization parameters and analysis conditions by evaluating the efficiency of three matrices: α-cyano-4-hydroxycinnamic acid, 2,5-dihydroxybenzoic acid (DHB), and 2-mercaptobenzothiazole in MALDI MS analysis. All results were compared to ESI MS (electrospray ionization mass spectrometry), and afterward, MALDI MSI analysis was performed on the leaf surface. This study showed through phytochemical analysis that G. sepium leaves are composed of polyphenols and tannins, concluding that the methanolic extract had a higher amount of flavonoid content. Four compounds were identified on the leaf surface, and their spatial distribution was analyzed by MALDI MS using DHB as a matrix. Kaempferol, isorhamnetin, and some fatty acids showed potential applicability for cosmetical use. All the extracts presented antioxidant activity or antimicrobial action and no cytotoxicity. Therefore, extracts of G. sepium could be used as raw materials in cosmetics.

Resumo-A espécie Macroptilium lathyroides (feijão-de-rôla) foi encontrada infestando áreas relativamente extensas de cultivo de soja na região Sul do Mato Grosso do Sul, onde produtores se queixaram da falta de eficiência de controle de herbicidas. Não existe nenhum herbicida registrado para o controle da espécie, e o número de pesquisas envolvendo a suscetibilidade a diversos princípios ativos é limitado. Objetivou-se com este estudo verificar a eficiência do herbicida glyphosate, isolado ou em associação a outros princípios ativos, no controle de M. lathyroides em três estádios de desenvolvimento, bem como encontrar outras alternativas de controle químico para utilização em pré e pós-emergência. O estudo foi instalado em casa de vegetação, constituindo-se do estabelecimento das plantas em vasos plásticos com 4 dm 3 de solo, onde herbicidas foram aplicados, conforme tratamentos, em três momentos: pré-emergência (4 dias após semeadura), pós-emergência inicial (4 a 6 folhas definitivas) e pós-emergência tardia (início do florescimento). Em pré-emergência, o M. lathyroides mostrou-se suscetível ao sulfentrazone e ao diuron; em pós-emergência inicial, glyphosate e trifloxysulfurom-sodium foram eficientes, e em pós-emergência tardia o glyphosate e o amonio-glufosinate também proporcionaram controle eficiente. A eficiência do carfentrazone-ethyl foi dependente do estádio de desenvolvimento da espécie e o pyrithiobac-sodium não demonstrou controle satisfatório. Novas avaliações devem ser conduzidas em condições de campo para confirmar os resultados obtidos neste estudo em ambiente controlado.
