Genetic diversity among proso millet (Panicum miliaceum) biotypes assessed by AFLP technique

Abstract: -The Amplified Fragment Length Polymorphism (AFLP) technique was used to access genetic diversity between three domestic and nine wild proso millet biotypes from the United States and Canada. Eight primer combinations detected 39 polymorphic DNA fragments, with the genetic distance estimates among biotypes ranging from 0.02 to 0.04. Colorado-Weld County black seeded and Wyoming-Platte County were the most distinct biotypes according to the dissimilarity level. A UPGMA cluster analysis revealed two distinct gro… Show more

“…A very limited number of DNA markers are reported in proso millet. A few RAPD and AFLP markers were reported in proso millet for genetic diversity analysis [10][11][12]. Among all PCR-based markers, microsatellites or simple sequence repeat (SSR) markers are the most informative and useful because they are co-dominant, multi-allelic, highly reproducible, abundant and are evenly distributed in the plant genome [13,14].…”

“…The results revealed extensive polymorphism among the accessions. A cluster dendrogram produced by similarity coefficient analysis revealed geographical groupings consistent with those of the M'Ribu and Hilu (1994) study, but not with the results of Karam et al (2004) described below. Hu et al (2008) mentioned "hard" (which millet breeders call non-glutinous) and "soft" (glutinous) grain characteristics among the samples.…”

“…This is not a distinction used widely in the literature, but is useful for interpreting how the various millet forms would be used and eaten [see section 3(b)]. Karam et al (2004) used the Amplified Fragment Length Polymorphism (AFLP) technique on three domestic and nine wild biotypes of proso millet. Subsequent cluster analysis resulted in two distinct groups without any geographical association.…”
