Mecanismos da intoxicação do fígado de rato causada pelo gossipol

Abstract: The liver plays a central role in metabolism due to its interposition between the digestive tract and the general circulation of the organism. It is also the main organ involved in biotransformation of exogenous substances (xenobiotics), with ability to convert hydrophobic compounds in water-soluble, more easily eliminated by the body. Gossypol is a toxic phenolic substance present in cotton seed (Gossypium sp.). Aiming to study the mechanisms involved in the hepatotoxicity of gossypol we evaluate its effects … Show more

“…and observed increases in GSH and reductions in GSSG concentration. However, the results obtained in the present study are consistent with those found by Carvalho et al [ 30 ] who observed a significant decrease in the level of GSH in liver homogenate of rats treated with 5 mg/kg BW gossypol compared to control animals, with a consequent increase in the concentration of GSSG. In addition, according to the results of the present study, vitamin E was able to protect against the oxidative effect of gossypol on glutathione.…”

“…These effects are consistently illustrated in this study, since there was an increase in the formation of MDA in the testis homogenate of animals treated with gossypol, indicating the occurrence of lipid peroxidation. A similar result was reported by Carvalho et al [ 30 ] when evaluating the effects of gossypol on rat livers. Treatment with vitamin E protected against this effect in the testes, which can be attributed to the fact that vitamin E is the major antioxidant in cell membranes, acting on polyunsaturated fatty acids and preventing their oxidation [ 34 ].…”

“…Even the liver being the main metabolizing organ of xenobiotics (De Carvalho et al, 2013), it was demonstrated that NDGA can cause cystic nephropathy in rats. Its occur due to the quinone resulting from the metabolism process of NDGA absorbed by the proximal tubular epithelium and accumulate in the lysosomes, thus leading to the destruction of tubular epithelial cells and tubule blockage (Goodman et al 1970;Evan, Gardner, 1979) (Figure 3).…”
