The Colletotrichum genus presents large genetic variability, as demonstrated by the occurrence of several pathogenic races and phenotypic traits. The objective of this study was to characterize 22 strains of C. lindemuthianum and Colletotrichum spp recovered from anthracnose lesions and bean scab, and to verify the relationship between species of the Colletotrichum genus, which inhabit anthracnose and scab lesions. Colony morphology, conidium size, the presence of septa, germination, sporulation, and mycelium growth rates, were analyzed in addition to the presence of mating-type genes, IRAP markers, and pathogenicity. Strains of Colletotrichum spp presented wide variation for all evaluated traits, indicating the presence of different species. Pathogenicity tests verified that the severity of the disease caused by strains of Colletotrichum spp must be evaluated 17 days after inoculation. Molecular analysis showed that only the C. lindemuthianum strains were grouped by the IRAP markers. For the physiological traits, we observed that C. lindemuthianum mycelium growth is slower than that of Colletotrichum spp strains. The information generated in this study confirms variability in the evaluated species of Colletotrichum and may direct future basic and applied studies aiming to control these diseases in common bean.

Common bean anthracnose is one of the main fungal diseases that affect the crop. The disease is caused by the fungus Colletotrichum lindemuthianum, anamorph, which has wide variability as well as its teleomorph. Sexual reproduction is the main mechanism of increasing genetic variability in fungi, therefore the study of morphological characteristics and their sexual structures are important. Homothallic strains of the pathogen have shown the production of new ascospores via mitosis after the sexual cycle. As a result of each mitotic division there are two ascospores with amygdaliform and/or allantoid morphology. In this work the characterization of this phenomenon is described as budding. This is the first report on the occurrence of ascospore budding in the genus Colletotrichum. The homothallic strain UFLA84‐1 was used. Analyses of light microscopy, fluorescence and high‐resolution scanning electron microscopy were used for the description of the budding process. Cytological analyses were carried out to estimate the dimension of ascospores, budding percentage and colony type. The budding cycle is described in six stages, and the complete process develops after the release of all ascospores from an ascus. The ascospores from this process can present both morphologies and bud growth can be apical or lateral. Sexual reproduction is a mechanism of genetic recombination and the budding process increases the number of ascospores generated by meiosis and may therefore represent an adaptive advantage of the pathogen.

The limit of the Colletotrichum gloeosporioides species complex is defined genetically, based on a strongly supported clade within the Colletotrichum ITS gene tree. All taxa accepted within this clade are morphologically more or less typical of the broadly defined C. gloeosporioides, as it has been applied in the literature for the past 50 years. We accept 22 species plus one subspecies within the C. gloeosporioides complex. These include C. asianum, C. cordylinicola, C. fructicola, C. gloeosporioides, C. horii, C. kahawae subsp. kahawae, C. musae, C. nupharicola, C. psidii, C. siamense, C. theobromicola, C. tropicale, and C. xanthorrhoeae, along with the taxa described here as new, C. aenigma, C. aeschynomenes, C. alatae, C. alienum, C. aotearoa, C. clidemiae, C. kahawae subsp. ciggaro, C. salsolae, and C. ti, plus the nom. nov. C. queenslandicum (for C. gloeosporioides var. minus). All of the taxa are defined genetically on the basis of multi-gene phylogenies. Brief morphological descriptions are provided for species where no modern description is available. Many of the species are unable to be reliably distinguished using ITS, the official barcoding gene for fungi. Particularly problematic are a set of species genetically close to C. musae and another set of species genetically close to C. kahawae, referred to here as the Musae clade and the Kahawae clade, respectively. Each clade contains several species that are phylogenetically well supported in multi-gene analyses, but within the clades branch lengths are short because of the small number of phylogenetically informative characters, and in a few cases individual gene trees are incongruent. Some single genes or combinations of genes, such as glyceraldehyde-3-phosphate dehydrogenase and glutamine synthetase, can be used to reliably distinguish most taxa and will need to be developed as secondary barcodes for species level identification, which is important because many of these fungi are of biosecurity significance. In addition to the accepted species, notes are provided for names where a possible close relationship with C. gloeosporioides sensu lato has been suggested in the recent literature, along with all subspecific taxa and formae speciales within C. gloeosporioides and its putative teleomorph Glomerella cingulata.Taxonomic novelties:Name replacement - C. queenslandicum B.
