Development of a UV/Vis spectrophotometric method for analysis of total polyphenols from Caesalpinia peltophoroides Benth

Abstract: Recebido em 23/8/11; aceito em 3/11/11; publicado na web em 4/1/12Caesalpinia peltophoroides is a domesticated tree found in Brazil. It was necessary to develop an analytical method to determine the content of total polyphenols (TP) in this herbal drug. The pre-analytical method was standardized for analysis time, wavelength, and the best standard to use. The optimum conditions were: pyrogallol, 760 nm, and 30 min respectively. Under these conditions, validation by UV/Vis spectrophotometry proved to be reliabl… Show more

“…In this case, the specificity was evaluated by the absence of matrix effects, considering that natural products are complex matrices in which it is not possible to obtain a pure analyte. The absence of matrix effects is verified if the curves of linearity and specificity are parallel, in other words, if both curves show the same or very similar slopes (Blainski et al, 2013;Bueno et al, 2012;Ribani et al, 2004). The data obtained in the linearity and specificity tests for F2 and TG appear in Table 2 and are represented in Figure 1.…”

“…The analytical conditions for UV/Vis spectrophotometric method were evaluated as described by Blainski et al (2013) and Bueno et al (2012), and were confirmed with the use of pyrogallol, for 30 min and 760 nm, respectively; reference substance, time reaction and wavelength, as described in material and methods.…”

“…A precisão é considerada uma das análises mais representativas, visto que revela o efeito das variações relacionadas aos dias, analistas ou equipamentos distintos, implicando garantia da reprodutibilidade do método (BUENO, 2012). A exatidão, por sua vez, reflete a proximidade dos resultados obtidos por uma metodologia analítica em relação aos valores verdadeiros (BRASIL, 2003).…”

“…The concentration of total Phenolics in the acetone leaf extracts of Sida cardifolia, Phyllanthus amarus and Adathoda vasica was determined by the Folin-Ciocalteu assay that involves reduction of the reagent by phenolic compounds, with concomitant formation of a blue complex, its intensity at 725nm increases linearly with the concentration of phenolics in the reaction medium 8 . The phenolic contents of the extracts were determined from calibration curve and were expressed in mg gallic acid equivalent/g of extract powder.…”
