Proteases de Leishmania: novos alvos para o desenvolvimento racional de fármacos

Abstract: Recebido em 7/6/09; aceito em 22/3/10; publicado na web em 29/6/10 Leishmania PROTEASES: NEW TARGETS FOR RATIONAL DRUG DEVELOPMENT. Leishmania causes tegumental and visceral diseases called leishmaniasis. Disease control is possible interrupting the transmission cycle, but HIV co-infection, chemotheraphy toxicity and lack of a vaccine are paramount difficulties. So, is necessary to study new Leishmania molecules and investigate the possibility to develop rational drugs using these molecules as targets. Leishma… Show more

“…These enzymes play a role in reducing viability and induction of morphological changes [85,86]. Roles of other enzyme systems include but limited to roles in metabolic activities like glycolysis, oxidation of fatty acids, lipid biosynthesis and purine salvage pathways [87][88][89][90][91][92][93][94][95][96][97][98][99][100][101][102][103]. Other Functions of the above mentioned biochemical and metabolic pathways include: Cell division cycle, transcription, apoptosis, cell proliferation, cell differentiation and innate immunity to activation of adaptive immunity [104][105][106][107][108][109][110][111][112][113].…”

“…These enzymes can be divided into endopeptidases and exopeptidases, depending on the site of hydrolysis. If the residue present at the catalytic site is taken into account, they are divided into metalloproteases, serine proteases, aspartic proteases and cysteine proteases [8]. In the genus Leishmania , cysteine proteases are the most abundant class of enzymes and are concentrated inside megasomes, where they act towards regulation of metabolic routes, cell differentiation in vertebrates and vectors, cell invasion and transposition of tissue barriers, degradation of hemoglobin and other hematopoietic proteins, evasion of the immune response, activation of inflammatory responses and programmed cell death [9].…”
