Characterization of resistance response to pathogens is a fundamental strategy in plant breeding programmes. Thus, in this study, we developed an effective inoculation method for resistance tests of bacterial halo blight (BHB), caused by Pseudomonas syringae pv. garcae on coffee. Firstly, aggressiveness of eight selected bacterial strains as well as seven mixed strains was assessed on coffee seedlings of Mundo Novo IAC 376‐4 cultivar. Two experiments were conducted comparing three inoculation techniques: initially, we compare the methods of sprinkling and multiple needles on four cultivars of Coffea arabica; later, four cultivars and seven genotypes were compared for the multiple needles method and the abrasion technique. Severity was evaluated according a disease rating scale (DRS), considering either leaf surface area affected for sprinkling, or affected inoculated area for inoculation by multiple needles and abrasion. The area under the disease progress curve of disease (AUDPC) was calculated considering weekly evaluations of disease from seven until 42 days postinoculation (DPI). The standard deviation (SD), coefficient of variation (CV) and confidence interval (CI) variables were used to denote the effectiveness of the methods. According to the results, strain IBSBF 1197 was the most aggressive, and three other strains showed high aggressiveness. All inoculation methods were able to discriminate the resistance response to BHB, wherein the sprinkling method was less efficient than multiple needles and abrasion technique was more efficient than multiple needles. In addition, disease evaluations at 14 DPI showed a high correlation coefficient with area under AUDPC at 42 DPI, validating the early selection to BHB. Results of inoculation methods indicated the abrasion on first pair of leaves, together with evaluations on fourteenth DPI, as the most promising technique for early selection on coffee breeding to BHB.

Twenty-five strains of Xanthomonas axonopodis pv. citri and 14 strains of Xanthomonas spp. were tested for bacteriocin production. X. axonopodis pv. passiflorae strains were sensitive to the bacteriocins produced by the 25 X. axonopodis pv. citri strains evaluated in this study while strains of X. axonopodis pv. manihotis and X. campestris pv. campestris showed variable sensitivity. Only five of the 25 X. axonopodis pv. citri strains were not inhibited by the bacteriocins produced by the two X. axonopodis pv. passiflorae strains. The bacteriocins produced by the Xanthomonas axonopodis pv. citri (FDC-806) and X. axonopodis pv. passiflorae (Mar-2850 A) strains were thermolabile, resistant to lysozyme and sensitive to DNAse. The bacteriocin produced by X. axonopodis pv. passiflorae was resistant to the action of proteinase K, trypsin and RNAse while the bacteriocin produced by X. axonopodis pv. citri was sensitive to these enzymes. The bacteriocins produced by X. axonopodis pv. passiflorae and X. axonopodis pv. citri were called passifloricin and citricin, respectively.

Avaliou-se a ocorrência da murcha-de-curtobacterium em lavouras de feijoeiro comum em algumas localidades do Estado de Santa Catarina, nas safras 2002/03 e 2003/04, e o comportamento dos genótipos BRS Valente, Carioca, CHC 97-29, CHP 97-26, CNPF 8104, Diamante Negro, Empasc 201 - Chapecó, IAPAR 44, IPR Graúna, IPR Juriti, IPR Uirapuru, LP 9728, Pérola, SCS 202-Guará, Sel. CP 9310635, TPS Bionobre, TPS Bonito, TPS Magnífico, TPS Nobre, TPS Soberano e Xamego perante Curtobacterium flaccumfaciens pv. flaccumfaciens (Cff), em condições de casa-de-vegetação. As cultivares IAC Carioca Akytã, IAC Carioca Aruã e IAC Carioca Pyatã foram empregadas como padrões de resistência a Cff. As avaliações dos sintomas ocorreram aos 5, 10, 15, 20 e 25 dias após a inoculação (DAI) e, posteriormente, foi estimada a área abaixo da curva de progresso da murcha-de-curtobacterium (AACPMC), em cada genótipo. A doença esteve presente nos municípios de Campos Novos, Faxinal dos Guedes, Guatambu, Ipuaçu, Ponte Serrada e Tigrinhos e que, aos 10 DAI, as cultivares SCS 202 - Guará e IPR Juriti mostraram baixa severidade. Porém, aos 25 DAI, somente as cultivares padrões foram resistentes e apresentaram menor AACPMC.
