Molecular diagnosis of eosinophilic meningitis due to Angiostrongylus cantonensis (Nematoda: Metastrongyloidea) by polymerase chain reaction-DNA sequencing of cerebrospinal fluids of patients

Abstract: Cerebrospinal fluid (CSF) samples from clinically diagnosed patients with detectableAngiostrongylus cantonensis-specific antibodies (n = 10), patients with clinically suspected cases that tested negative for A. cantonensis-antibodies (n = 5) and patients with cerebral gnathostomiasis (n = 2) and neurocysticercosis (n = 2) were examined by a single-step polymerase chain reaction (PCR)

“…A small study in Thailand indeed found Angiostrongylus DNA in CSF from four of 10 serologically confirmed angiostrongyliasis cases using a genus-level standard PCR. 22 The aim of this work was to evaluate if a species-specific real-time PCR assay, originally developed to detect A. cantonensis in host animals, 23 could be suitable for the detection of parasite DNA in CSF specimens from patients with CNS angiostrongyliasis.…”

“…The peak concentration of eosinophils in both CSF and peripheral serum has been reported at DAI 25–35 [ 8 ]. Immunological methods and PCR, such as ELISA, dot immunogold filtration assay (DIGFA) and PCR based on the 18S rRNA, were successfully applied for the identification of A. cantonensis [ 11 – 13 ]. However, positive results of ELISA or DIGFA can hardly be obtained in the acute phase of the disease.…”

“…La meningitis eosinofílica causada por A. cantonensis se diagnostica en gran medida a partir de datos epidemiológicos y de la sospecha clínica, lo que es posible cuando el personal está entrenado y alertado. Para el diagnóstico en el laboratorio se han desarrollado algunas PCR de poca sensibilidad y resultados contradictorios, pruebas que no están disponibles en el comercio (15,24,(27)(28)(29)(30)(31)(32)(33)(34)(35).…”
