BackgroundHuman and animal fascioliasis is emerging in many world regions, among which Andean countries constitute the largest regional hot spot and Peru the country presenting more human endemic areas. A survey was undertaken on the lymnaeid snails inhabiting the hyperendemic area of Cajamarca, where human prevalences are the highest known among the areas presenting a "valley transmission pattern", to establish which species are present, genetically characterise their populations by comparison with other human endemic areas, and discuss which ones have transmission capacity and their potential implications with human and animal infection.MethodsTherefore, ribosomal DNA ITS-2 and ITS-1, and mitochondrial DNA 16S and cox1 were sequenced by the dideoxy chain-termination method.ResultsResults indicate the presence of three, morphologically similar, small lymnaeid species belonging to the Galba/Fossaria group: Galba truncatula, Lymnaea neotropica and L. schirazensis. Only one combined haplotype for each species was found. The ITS-1, 16S and cox1 haplotypes of G. truncatula are new. No new haplotypes were found in the other two species. This scenario changes previous knowledge, in which only L. viator (= L. viatrix) was mentioned. Galba truncatula appears to be the most abundant, with high population densities and evident anthropophyly including usual presence in human neighbourhood. Infection by Fasciola hepatica larval stages were molecularly confirmed in two populations of this species. The nearness between G. truncatula populations presenting liver fluke infection and both human settings and schools for children, together with the absence of populations of other lymnaeid species in the locality, suggest a direct relationship with human infection.ConclusionsThe geographical overlap of three lymnaeid species poses problems for epidemiological studies and control action. First, a problem in classifying lymnaeid specimens in both field and laboratory activities, given their transmission capacity differences: G. truncatula mainly involved in transmission to humans, L neotropica typically responsible for livestock infection, and L. schirazensis unable for transmission. Although several phenotypic characteristics may be helpful for a preliminary specimen classification, a definitive classification can only be obtained by marker sequencing. Aditionally, L. schirazensis increases the confusion, owing to its ability to mix with other Galba/Fossaria species and distort fascioliasis data such as transmission capacity and infection susceptibility. Second, a problem for epidemiological analysis, surveillance and control by methods as mathematical modelling and Remote Sensing - Geographical Information Systems. In Cajamarca, low resolution mapping may be insufficient, as already verified in Andean areas where different lymnaeid species overlap.

We examined the patelliform snails of the subfamily Lancinae, endemic to northwestern North America, to test whether morphological variation correlated with genetic and anatomical differences. Molecular analyses using cox1, 16S, calmodulin intron, and 28S rDNA partial sequences and anatomical data supported recognition of four species in three genera. The relationships of lancines within Lymnaeidae are not yet well-resolved. The federally endangered Banbury Springs lanx is described as a new genus and species, Idaholanx fresti, confirming its distinctiveness and narrow endemicity.

According to Brown (1978) and Mas-Coma et al. (2005), members within this family can sustain larval development of more than 70 different trematodes. As epidemiological studies made on these parasites need the accurate identification of the snail host, several methods using shell morphology and/or the anatomy of the reproductive system have been proposed over the years to classify these lymnaeids (Hubendick, 1951; Burch, 1982).Shell characters have been extensively, and very often exclusively, used to build the systematics of molluscs. Marine shells are diversified and present a very large set of morphological characters, i.e. protoconch structure, teleoconch ornamentation, radula morphology, etc…, that have proved useful in malacological taxonomy. In freshwater pulmonates, these shell characters are for the most part absent and shell shape only has been used to describe new taxa in the last 19 th and 20 th centuries and even sometimes in most recent years. As a result of using shell as diagnostic species character, a plethora of names may be found in the literature on freshwater pulmonates, numerous probably being synonyms. The case of the Lymnaeidae family is caricatural with about 1200 described species and several dozens of genera (Hubendick, 1951; Burch, 1982), whereas recent studies suggested that the family contained approximately 100 species (Strong et al., 2008; Jarne et al., 2010).The classification systems based on morphological characters of shell shape have rapidly generated a controversy because lymnaeids exhibited a great diversity in shell morphology linked to substantial ecophenotypic plasticity (Samadi et al., 2000; Hurtrez-Boussès et al., 2005;Schniebs et al., 2011). It is accepted that anatomical characteristics of the snail's reproductive system are more homogeneous (Pointier et al., 2009; Correa et al., 2011), but only DNA-based analyses, i.e. phylogeny and barcoding, developed from the 2000s could effectively solve this taxonomic problem. As these last analyses allowed to ascribe individuals to one species or another, several new species such as Lymnaea neotropica (Bargues et al., 2007), Galba sp. (Correa et al., 2010(Correa et al., , 2011 or Lymnaea schirazensis (Bargues et al., 2011) have been recognized and/or described.The conchological characteristics of Galba sp. correspond to that of several dozens of taxa reported in North America, presumably the geographic origin of this small-shelled species clade (see Hubendick, 1951; Burch, 1982; Correa et al., 2010; Johnson et al., 2013).
