Evidences of gentamicin resistance amplification in Klebsiella pneumoniae isolated from faeces of hospitalized newborns

Abstract: The intestinal microbiota, a barrier to the establishment of pathogenic bacteria, is also an impor using as a probe a labeled cloned insert containing aminoglycoside modifying enzyme (AME) gene sequence originated from a plasmid of a Klebsiella pneumoniae strain previously isolated in the same hospital. Further, we found indications of inactivation to other resistance genes in variants selected under similar conditions, as well as, indications of co-amplification of other AME markers (amikacin).Since the intes… Show more

“…Some other studies have shown that Amx would enrich the betalactam resistance genes and corresponding antibiotic resistant bacteria in the intestine [39,54]. Taking Gen would also promote the spread of antibiotic resistant bacteria and ARGs [15,16]. The above conclusions would help us infer the combined exposure effects on the increase of ARGs and human disease-related genes in this study.…”

“…Besides, gentamicin (Gen) is known as an aminoglycoside drug that binds to the 30S subunit of the ribosome to block bacterial protein synthesis [14]. Over the last several years, the effects of Amx or Gen on the human intestinal microbiota have been extensively studied [15][16][17][18][19]. However, very few research papers have reported their combination effects on intestinal microbial ecosystem.…”

“…Immunosuppression, stress, administration of antibiotics or antacids play an important role in altering defense mechanisms, allowing microorganisms present in food to replace the normal gastrointestinal £ora. Additionally, the intestinal environment is a scenario of selective processes due to therapeutic and prophylactic use of antimicrobial agents representing an imperceptible reservoir of multiresistant bacteria [3]. “…The importance of the colonization of skin and mucosa has been recognized as a crucial step in the development of nosocomial infections [2]. Additionally, the intestinal environment is a scenario of selective processes due to therapeutic and prophylactic use of antimicrobial agents representing an imperceptible reservoir of multiresistant bacteria [3].…”

“…In a previous study with Enterobacteriaceae strains of hospital origin we searched for genetic elements which express multidrug-resistance including resistance to aminoglycosides (10). Probes were built to detect plasmids encoding these multidrugresistance traits.…”
