An improved silver staining procedure for schizodeme analysis in polyacrylamide gradient gels

Abstract: A simple protocol is described for the silver staining of polyacrylamide gradient gels used for the separation of restriction fragments of kinetoplast DNA [schizodeme analysis of trypanosomatids (Morel et al., 1980)]. The method overcomes the problems of non-uniform staining and strong background color which are frequently encountered when conventional protocols for silver staining of linear gels are applied to gradient gels. The method described has proven to be of general applicability for DNA, RNA and prote… Show more

“…The samples were denatured prior to analysis by adding one μL of 0.5 M NaOH/10 mMEDTA and then electrophoresed for 3 h at 10 V/cm using 0.5 × TBE buffer (pH8.4). SSCP patterns were visualized by silver staining as described elsewhere [ 30 ].…”

“…The digestion products were electrophoresed on a 4.5-10% polyacrylamide gradient gel and stained with silver nitrate. 11 The quantification of common bands was per- formed with the formula C/A ϩ B ϩ C, where C is the number of bands common to the two stocks being compared, and B and A correspond to bands present only on the first and second stocks, respectively.…”

“…Soluble proteins (40 μg) were resolved using 12% sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and silver staining, as previously described [29,30]. Electrophoresis was performed at 25°C in a Mini Protean II system (BioRad Laboratories, US).…”
