Introduction: Governador Valadares is an endemic area of American tegumentary leishmaniasis (ATL). The detection rate was 15.36 per 100,000 habitants from to 2006(Miranda, 2008. This study aimed to analyze the effects of age on the frequency of adverse reactions caused by antimony in the treatment of ATL in the City of Governador Valadares, State of Minas Gerais, Brazil, during 2009. Methods: Data were collected from the forms of the Information System for Notifiable Diseases, and from charts, questionnaires, and home visits to patients. Results: The study included 40 patients, 26 (65%) of whom were males. Individuals over the age of 50 had a 66% higher rate of adverse effects than subjects who were 50 years old or less (CI 95%, 1.14-2.41). The average age of individuals who reported some type of adverse effect was 44.11 years (SD = 20.14), while the average age of the group that did not report any adverse effect was of 25.46 years (SD = 18.37; p < 0.01). Clinical healing was 67.5%, and 10% of patients discontinued the treatment. Conclusions: In the treatment of ATL, the age of patients should be considered, because most adverse reactions occur in individuals over 50 years of age. For this reason, the drug should be used with restriction in these cases.

Visceral leishmaniasis is caused by parasites of the genus Leishmania. Prevalent in developing countries, the disease is on the list of the world's most neglected diseases. Most cases (90%) in Latin America occur in Brazil, especially in the Northeast. The condition is treated mainly with pentavalent antimonials, an expensive and relatively inefficient drug known to induce resistance. In search of new antileishmanial drugs, much attention has been given to the medicinal properties of coumarin. In this study, coumarin was isolated from seeds of Amburana cearensis, purified and evaluated with regard to its ability to inhibit Leishmania chagasi promastigotes using the MTT test. The cell viability of macrophages treated with coumarin was also evaluated. The findings were submitted to one-way ANOVA for paired data, followed by the Bonferroni correction. The level of statistical significance was set at 5% (p < 0.05). Coumarin displayed low toxicity to macrophages in the MTT test (p > 0.05), but was toxic to Leishmania chagasi promastigotes (p < 0.05). Our results represent a contribution to the development of new drugs for the control or prophylaxis of visceral leishmaniasis.

RESUMOA exposição a agentes químicos durante a prenhez pode alterar o desenvolvimento animal e a maturação dos sistemas fisiológicos. O Antimoniato de Meglumina (AM) é a primeira escolha no tratamento das leishmanioses, mesmo apresentando toxicidade e suposta impregnação neural. Objetivo: O objetivo desde estudo foi verificar o desenvolvimento físico da prole de camundongos Swiss, expostos ao AM durante o período fetal. Material e método: Fêmeas prenhes foram tratadas com AM (100mg/kg/dia) durante o 13° ao 18° dia de gestação -ddg e o grupo controle foi tratado com o veículo. No dia zero pós-natal -dpn foi registrado o número e peso da prole de cada ninhada e avaliou-se o desenvolvimento físico (nascimento de pêlos, abertura de olhos e orelhas, dente inciso, abertura da vagina e descida dos testículos) do 0 ao 31º dpn. Os filhotes foram pesados no 3°, 5°, 7°, 14° e 21° dpn. Resultados: Não houve diferenças significativas no número e peso da prole no 0 dpn entre os grupos experimental e controle. O desenvolvimento ponderal da prole de ambos os grupos foi semelhante, assim como o aparecimento dos indicadores de desenvolvimento físico. Conclusão: A exposição ao AM durante o período fetal em camundongos não interferiu no desenvolvimento físico das crias. DESCRITORESLeishmaniose. Antimônio. Meglumina. Toxicidade de Drogas. SUMMARYThe exposition to the quimical agents during the pregnant can alter the animal development and the maturation of the physiological systems. The Meglumine antimoniate (MA) is the first-choice of treatment of of leishmaniosis same presented toxicity and suppond neural impregnation. Objective: The aim this work was verify the physical developmental of the pups of Swiss mice exposed to MA during the fetal period. Material and methods: Pregnant dams received MA (100mg/kg/day) during day 13° to 18° of gestation -ddg. Control group received deionized water. At birth day (0 postnatal -dpn) the number and corporal weight of the pups of each litter was registered. The corporal weight of the pups was realized on day 3°, 5°, 7°, 14° and 21° postnatal. Physical development was assessed by monitoring daily on the 0 to 31º dpn the following parameters in 16 litters: birt of pilus, eye opening, ear unfolding, incisor eruption, vagina opening and descent of the testis. Results: No statistic difference was observed in the number and corporal weight between the experimental and control pups at 0 dpn. Conclusion: The ponderal development of the pups of both the groups was similar, as well the appearance of indicators of the physical development. The maternal exposition in mice to MA throughout the fetal period did not interfere in the physical development of the pups. DESCRIPTORS Leishmaniasis. Antimony. Meglumine.
