Evaluation of dengue NS1 antigen detection for diagnosis in public health laboratories, São Paulo State, 2009

Abstract: SUMMARYThe present work evaluated the diagnostic accuracy of detection of Dengue NS1 antigen employing two NS1 assays, an immunochromatographic assay and ELISA, in the diagnostic routine of Public Health laboratories. The results obtained with NS1 assay were compared with virus isolation and, in a subpopulation of cases, they were compared with the IgM-ELISA results obtained with convalescent samples. A total of 2,321 sera samples were analyzed by one of two NS1 techniques from March to October 2009. The sampl… Show more

“…Accurate diagnosis of dengue can be confirmed based on serum positivity for NS1 antigen, IgM, and IgG antibodies. The NS1 antigen can be determined by ELISA, which is highly sensitive, specific, 12 and helpful in the early diagnosis of acute infection. In our study, we observed that platelet count determination had been used in most (50%) of the cases to diagnose dengue infection.…”

“…10 It is available in both district and reference centres. 4 According to Bisordi et al, 11 the NS1 assay is highly suitable for samples undergoing screening for virus isolation. Study of Andries et al, 12 also revealed that the discovery of the NS1 protein as an early marker for DENV infection, especially in rapid diagnostic test format now allows dengue diagnosis during the early phase of the disease, even in laboratories with limited equipment and human resources.…”

“…Moreover, among the studies that were included in this review, only Dos Santos et al, used the ELISA to capture the NS1 antigen in the acute phase of dengue [45]. This method is more sensitive than traditional RT-PCR and is used in many laboratories for the early diagnosis of dengue [65].…”
