Diagnosis of pulmonary pneumocystosis by microscopy on wet mount preparations

Abstract: SUMMARYWe have compared the searching of the presence of "honeycomb" structures by direct microscopy on wet mount preparations with the direct immunofluorescence (DIF) for the diagnosis of Pneumocystis carinii pneumonia (PCP) in 115 bronchoalveolar (BAL) fluids. The samples belonged to 115 AIDS patients; 87 with presumptive diagnosis of PCP and 28 with presumptive diagnosis other than PCP.The obtained results were coincident in 114 out of 115 studied samples (27 were positive and 87 negative) with both techniq… Show more

“…A study of 389 children at risk for or infected with HIV-1 conducted from February 1990 to June 1997 found that severe bacterial infection and PcP were the most common AIDS-defining conditions and death-related diseases among AIDS patients (Fallo et al , 2002). Another study demonstrated a high rate of PcP (35%) among AIDS patients in intensive care units (Bava et al , 2002). Although the comparison of the pre and post-HAART era in Argentine AIDS patients revealed an increased PcP rates from 5.9% to 9.4%, TB was the main cause of hospital admission for HIV patients in both study periods (Pérez et al , 2005).…”

“…The sample was centrifuged (1 500 rpm/15´), and fresh microscopy, smears stained with Kinyoun [4] , Giemsa (Figure 1) and a rapid modification of the Grocott technique [5] were performed. Also, in our center the number of pulmonary cryptococcosis diagnosed in AIDS patients is significantly lower than expected as compared with the cases of meningoencephalitis which is produced by the infection of Cryptococcus neoformans (C. neoformans) through inhalation of the propagules [6] . Part of the respiratory secretions was processed in the microbacteriology, mycology and bacteriology services of the Muñiz Hospital and the result was negative.…”

“…Part of the respiratory secretions was processed in the microbacteriology, mycology and bacteriology services of the Muñiz Hospital and the result was negative. The wet smear revealed numerous leukocytes, filariform and rhabditiform larvae of S. stercoralis and foamy exudates [6] . A quick modification of the Grocott technique to the smears showed P. jiroveci cysts, while the Kinyoun stain was negative for acid resistant bacteria and protozoa.…”
