The present study developed Galleria mellonella and murine infection models for the study of Trichosporon infections. The utility of the developed animal models was demonstrated through the assessment of virulence and antifungal efficacy for 7 clinical isolates of Trichosporon asahii, T. asteroides and T. inkin. The susceptibility of the Trichosporon isolates to several common antifungal drugs was tested in vitro using the broth microdilution and the Etest methods. The E-test method depicted a lower minimal inhibitory concentration (MIC) for amphotericin and a slightly higher MIC for caspofungin, while MICs observed for the azoles were different but comparable between both methods. All three Trichosporon species established infection in both the G. mellonella and immunosuppressed murine models. Species and strain dependent differences were observed in both the G. mellonella and murine models. T. asahii was demonstrated to be more virulent than the other 2 species in both animal hosts. Significant differences in virulence were observed between strains for T. asteroides in the murine model. In both animal models, fluconazole and voriconazole were able to improve the survival of the animals compared to the untreated control groups infected with any of the 3 Trichosporon species. In G. mellonella, amphotericin was not able to reduce mortality in any of the 3 species. In contrast, amphotericin was able to reduce murine mortality in the T. asahii or T. inkin models, respectively. Hence, the developed animal infection models can be directly applicable to the future deeper investigation of the molecular determinants of Trichosporon virulence and antifungal resistance.

341 IntroduçãoPeritonite é complicação freqüente no tratamento da insuficiência renal crônica por diálise peritoneal ambulatorial continuada (CAPD). A maioria dos episódios de peritonite é causada por bactérias, em especial por estafilococos, mas um número crescente de peritonites causadas por fungos tem sido relatado 1,2 . Na maioria dos casos os agentes pertencem ao gênero Candida, mas espécies incomuns são associadas com maior freqüência aos episódios de peritonites fúngicas durante tratamento por CAPD. Relatamos um caso de peritonite por Trichosporon beigelii em criança com dois anos, durante tratamento por CAPD. Relato do casoPaciente do sexo masculino, branco, com dois anos de idade, nascido a termo de parto vaginal. Apresentava ausência total da musculatura abdominal, criptorquidia, hipoplasia pulmonar à esquerda, rim esquerdo hipoplásico e direito com hidronefrose, além de malformações ureterais. Com 10 dias de idade foi submetido à cirurgia para correção da criptorquidia, nefrectomia à direita e implante do catéter de Tenckhoff, iniciando programa de CAPD. Apresentou infecções respiratórias de repetição, hiperpotassemia persistente, três episódios de peritonite por Staphylococcus aureus tratados com vancomicina e rifampicina e um por Streptococcus sp., tratado com vancomicina e amicacina. Em 17.02.95, apresentou febre e efluente turvo, com celularidade aumentada (854/ul: 79% neutrófilos e 21% linfócitos). Embora o resultado negativo para bactérias no exame direto e cultivos, foi iniciado tratamento com vancomicina, persistindo os sintomas de peritonite. Exame micológico realizado em 21.02.95 evidenciou, no sedimento do efluente, fragmentos de hifas hialinas, artroconídios e blastoconídios. Cultivos em BHI ResumoRelatamos um caso de peritonite por Trichosporon beigelii em uma criança com dois anos de idade, durante tratamento por diálise peritoneal ambulatorial continuada (CAPD). O diagnóstico foi feito pelo exame direto e isolamento em cultivo do sedimento do dialisado. São comentadas as infecções na criança, causadas por este patógeno emergente.
