Absence of sibutramine effect on spontaneous anxiety in rats

Abstract: Introduction: Sibutramine has been described as a drug recommended for treatment of obesity, since it has the ability to inhibit the reuptake of serotonin and noradrenaline in the central nervous system, thereby increasing energy expenditure. Objective: Investigate the anxiogenic and anxiolytic effects of acute and chronic treatment with sibutramine in rats submitted to the task of the elevated plus-maze. Methods: Diazepam was used as a positive control for the anxiolytic effect, and the task of the elevated p… Show more

“…In this study, EPM results showed that rats in the diabetic groups exhibited more entries into the closed arms and time spent in the closed arms, which were reversed by loganin treatment. Frassetto et al (14) used diazepam (1mg/kg) as a positive control in their study for its anxiolytic effect and showed that administration of diazepam increased the percentage of entries and time spent in the open arms when compared with diabetic rats. This study has failed to demonstrate this result and there was no apparent anxiolytic activity of diazepam in diabetic rats in EPM.…”

“…We reviewed the available literature on diazepam for the three major rodent ARDEB assays: EPM, OF and LD. For the meta-analysis, data was Rodent anxiety meta-analyses page 11 extracted from a randomized subset of fifteen studies (out of 62 eligible papers) (Brioni et al, 1994;Faria et al, 1997;Boerngen-Lacerda and Souza-Formigoni, 2000;LaBuda and Fuchs, 2001;Hui et al, 2002;Chen et al, 2004;Jain et al, 2005;Jászberényi et al, 2009;Frassetto et al, 2010;Wanasuntronwong et al, 2012;Onusic et al, 2002;Birkett et al, 2011;Costa et al, 2011;Choleris et al, 2001;González-Pardo et al, 2006). Calculation of an average Hedges' g (Cumming, 2012) revealed that diazepam had a moderate effect on ARDEB, with a -0.89 g [95CI -1.12, -0.6] reduction compared with untreated control animals ( Figure 3).…”

“…Its mechanism of action – similarly to tricyclic antidepressants – involves the blockade of the neuronal reuptake of serotonin and noradrenaline . Considering that tricyclic antidepressants, such as imipramine, are employed for the treatment of pathologies such as generalized anxiety disorder and panic disorder in human beings , it was previously investigated whether sibutramine would alter both anxiety‐ and panic‐related behaviours displayed by male rats submitted to animal tests of anxiety . Sibutramine did not affect the behaviour of male rats in the elevated plus‐maze (EPM), but it decreased the inhibitory avoidance in both light–dark transition test and elevated T‐maze (ETM) and impaired the escape response in the ETM .…”
