Antiproliferative and apoptotic potencies of glucocorticoids: nonconcordance with their antiinflammatory and immunossuppressive properties

Abstract: Relative antiinflammatory and immunosuppressive potencies of glucocorticoids (GC) were previously well defined. Nonetheless, GC also regulate cell proliferation and programmed death (apoptosis). The aim of this study was to determine the relative potency of different GC on the modulation of cell survival. The GC-sensitive lymphoblast cell line CEM-c7/14 was submitted to 48 h-exposure to GC (dose-response curve from 10(-8) to 10(-5) M). Cell survival was analyzed employing the DimethylTiazol-Tetrazolium (MTT) t… Show more

“…Group 1 (n = 5) were injected with hydrocortisone (0.2 mg/200 l/mouse, NO 04050910, purchased from Jinyao Co., Tianjin, China) and cyclophosphamide (1 mg/200 l/mice, NO 0502221 Hengrui Co., Jiangsu, China) every second day for total three times (15,16) as immuno-compromised mouse. Group 2 (n = 5) were injected with only PBS as immunocompetent mice.…”

“…Glucocorticoids can modulate cell proliferation, 4 as they reduce the expressions of the heterodimer jun-fos and c-myc, among other transcription factors that decide on cell overlife and multiplication, determining the pause between G1 phase and S phase of the cell cycle. They can also induce apoptotic cell death, culminating in the activation of proteins with nuclear activity involved in the degradation of DNA, RNA and other structural cell proteins.…”
