Tuberous Sclerosis Complex, also known as Epiloia or Bourneville-Pringle disease is an autosomal dominant neurocutaneous syndrome with variable clinical expression. It is a multisystem disorder that may be associated with hamartomas in multiple organs in an unpredictable manner. The dermatologist plays an essential role in the history of the disease, since skin manifestations represent the most prevalent clinical features, enabling early diagnosis and intervention in its natural course. This article aims to inform the scientific community about advances made in the study of genetics and molecular biology. Recent findings regarding stimulation of tumor growth have been changing the history of this condition, making therapeutic trials with topical and systemic drugs possible. Knowledge of these topics enables better management of the patients affected, since tissue replacement by tumors can result in significant morbidity and mortality. Keywords: Dermatology; Diagnosis; Neurology; Sirolimus; Tuberous sclerosis Resumo:A Esclerose Tuberosa, também conhecida como Epilóia ou Facomatose de Pringle-Bourneville, é uma síndrome neurocutânea de caráter autossômico dominante com expressões clínicas variadas. É uma doença multissistêmica que pode cursar com hamartomas em diversos órgãos, de forma imprevisível. O dermatologista tem papel essencial na história da doença, uma vez que as afecções cutâneas representam as mais prevalentes apresentações clínicas, possibilitando assim o diagnóstico precoce da sín-drome e intervenção na sua evolução natural. O presente artigo tem o objetivo de atualizar a comunidade científica sobre avanços alcançados no estudo genético e biologia molecular. Recentes descobertas sobre o estímulo do crescimento tumoral vêm mudando a evolução desta patologia, possibilitando ensaios terapêuticos com drogas tópicas e sistêmicas. O conhecimento destes aspectos possibilita melhor condução dos pacientes acometidos, dado que a substituição tumoral dos diversos tecidos pode resultar em relevante morbidade e mortalidade.

Despite the constant advances in the treatment of cancer, this disease remains one of the main causes of mortality worldwide. So, the development of new treatment modalities is imperative. Snake venom causes a variety of biological effects because they constitute a complex mixture of substances as disintegrins, proteases (serine and metalo), phospholipases A2, L-amino acid oxidases and others. The goal of the present work is to evaluate a anti-tumor activity of some snake venoms fractions. There are several studies of components derived from snake venoms with this kind of activity. After fractionation of snake venoms of the families Viperidae and Elapidae, the fractions were assayed towards murine melanoma cell line B16-F10 and fibroblasts L929. The results showed that the fractions of venom of the snake Notechis ater niger had higher specificity and potential antitumor activity on B16-F10 cell line than the other studied venoms.Since the components of this venom are not explored yet coupled with the potential activity showed in this work, we decided to choose this venom to develop further studies. The cytotoxic fractions were evaluated to identify and characterize the components that showed antitumoral activity. Western blot assays and zymography suggests that these proteins do not belong to the class of metallo and serine proteinases.

Stress is a defense mechanism of an organism and is directly related to homeostasis, which is the equilibrium state of the multitude of organism systems, both among each other and with the environment. Given that stress is a major cause of cellular instability, it is directly related to its loss homeostasis. The present study assessed the changes caused by stress on the genetic material of Rattus norvegicus using micronuclei analysis. A total of 10 males were studied by being submitted to continuous stress from photoperiod, temperature and noise for 26 days. Peripheral blood samples were obtained immediately before submitting the animals to stress, as well as on the fifteenth and twenty‐sixth days afterwards. Blood smears were stained with Giemsa and the slides were analyzed using light microscopy. Approximately 3,000 polychromatic erythrocytes from each animal were observed and classified as either normal or micronucleated. The analysis indicated a frequency of 0.0012014 for the control sample, whereas samples on days 15 and 26 had frequencies of 0.0035458 and 0.0496850, respectively. A statistical analysis revealed statistically significant differences among the studied cell samples (ANOVA p < 0.00001, significance of 5%). Therefore, the presence of micronuclei in the studied samples is consistent with a cause/effect relationship. Copyright © 2010 John Wiley & Sons, Ltd.
