Combination of cafeteria diet with intraperitoneally streptozotocin in rats. A type-2 diabetes model

Abstract: To develop a model of induction of type-2 diabetes (DM2) by combining low doses of streptozotocin (STZ) and a cafeteria diet. Methods: Forty male Wistar rats (200 g) were allocated into four groups: control (non-diabetic, n = 10); STZ 30 mg/kg (diabetic, n = 10); STZ 35 mg/kg (diabetic, n = 10); and STZ 40 mg/kg (diabetic, n = 10). DM2 was induced with a single intraperitoneal injection of STZ after four weeks of cafeteria diet in the three diabetic groups. All animals were evaluated as for anthropometric, and… Show more

“…After 10 days, the animals received an intraperitoneal injection of streptozotocin (40 mg·kg –1 ; Sigma Chemical, St. Louis, MO, USA) dissolved in a 20 mmol·L –1 sodium citrate solution (pH = 4.5). The streptozotocin dosage performed in this study for the experimental model of type 2 diabetes used, simulates metabolic characteristics of DM2 in humans 16 . Blood glucose was measured 48 h after the injection, and fasting glycemia > 250 mg·dL –1 was considered hyperglycemia.…”

“…Intraglomerular pressure is primarily regulated through the classical angiotensin II pathway, where the renin-angiotensin system (RAS) activation leads to preferential vasoconstriction of the efferent arteriole. However, mounting evidence suggests the existence of numerous other angiotensin peptide products generated by endo-and carboxypeptidases, including angiotensin III (Ang 2-8), Ang 1-7, Ang 1-9, Ang 3-7, and Ang 3-8, each exerting distinct physiological effects [16,17]. Angiotensin II, a key player, increases intraglomerular pressure, a pivotal factor in CKD progression.…”
