Recent data show that young people, mainly due to the pressure of some risk factors or due to disrupted interpersonal relationships, utilise greater reward value and display greater sensitivity to the reinforcing properties of “pleasurable stimuli”, specifically in those situations in which an enhanced dopamine release is present. Alcoholic beverages, foods rich in sugar and fat, and illicit drug use are pleasurable feelings associated with rewards. Research shows that there is a link between substance abuse and obesity in brain functioning. Still, alcohol excess is central in leading to obesity and obesity-related morbidities, such as hepatic steatosis, mainly when associated with illicit drug dependence and negative eating behaviours in young people. It is ascertained that long-term drinking causes mental damage, similarly to drug abuse, but also affects liver function. Indeed, beyond the pharmacokinetic interactions of alcohol with drugs, occurring in the liver due to the same metabolic enzymes, there are also pharmacodynamic interactions of both substances in the CNS. To complicate matters, an important noxious effect of junk foods consists of inducing obesity and obesity-related NAFLD. In this review, we focus on some key mechanisms underlying the impact of these addictions on the liver, as well as those on the CNS.

A hepatite C é uma enfermidade que atinge o fígado causando sua inflamação, alcançando milhares de pessoas todos os anos, contudo atualmente ainda não existem vacinas para o vírus responsável pela doença, o HCV, embora estudos já sejam realizados sobre a viabilidade. Assim sendo, a vacinologia reversa pode ser de grande auxílio para detecção de alvos promissores para utilização como antígeno vacinal e concomitantemente formulação de uma vacina eficaz. O respectivo trabalho tem como objetivo obter dados relevantes para o desenho de vacinas promissoras contra o HCV através da utilização de ferramentas de bioinformática. Para tanto, após as sequências do vírus serem obtidas no Centro Nacional de Informação Biotecnológica (NCBI), foram realizadas as análises do potencial antigênico através do VaxiJen v.2.0. e do ANTIGENpro, juntamente foi verificado o potencial alergênico com o AlgPred, logo após as proteínas serem selecionadas, seguiriam para as próximas etapas onde foram submetidas para a caracterização das propriedades físico-químicas, através do ProtParam e a predição de epítopos de células T foi realizada no NetTepi. Dentre os resultados obtidos pode-se notar que, a proteína E2 foi a única a se demonstrar alergênica, já à proteína E1 não apresentou potencial antigénico e as demais proteínas foram classificadas como instáveis, seguindo somente a proteína NS4a para predição de epítopos de células T.

The aim is to investigate the prevalence, risk factors, and hepatitis C virus (HCV) genotypes/subtypes among crack users in‐treatment in Central Brazil. A cross‐sectional survey in which 600 in‐treatment crack users were interviewed and tested for anti‐HCV Ab by enzyme‐linked immunosorbent assay was conducted between August 2012 and April 2013. Anti‐HCV‐positive samples were also submitted for HCV RNA detection by polymerase chain reaction. Positive HCV RNA samples were genotyped by direct sequencing analysis of the NS5B region of the viral genome, followed by phylogenetic analysis. Of the total, 3.7% (95.0% CI, 2.4%‐5.6%) were anti‐HCV positive. Age over 40 years and history of injecting drugs were risk factors for HCV, while snorting cocaine was a protector variable. HCV RNA was detected in 14 of 22 anti‐HCV‐positive samples, and the genotypes 1 (n = 10) and 3 (n = 2), subtypes 1a (n = 7), 1b (n = 3), and 3a (n = 2) were identified. The HCV prevalence found among crack users is almost threefold that observed in the general population in Brazil supporting that this population is at higher risk for HCV. The findings of cocaine insufflation as a protective behavior for HCV infection in this population should be explored.
