Eucalyptus spp. it is known to be part of most forests planted for commercial use in Brazil, however when intercropped with Pitaia (Hylocereus undatus), few studies report this interaction. The objective of this work was to evaluate the effects of shading and the existence of allelopathic interaction between Eucalyptus spp and pitaia (H. undatus) in the field, as well as the action of eucalyptus leaf extract in the germination of pitaia seeds in laboratory conditions. In the field experiment, the statistical model of randomized block design (DBC) was used, with the use of adult plants of pitaia and eucalyptus, evaluating the following variables: number of shoots in the aerial part; number of flowers; crown diameter; circumference of the cladode; number of fruits and the variation in the amount of light reaching the plants. It was observed that the plants that were outside the eucalyptus forest showed better results. In the second experiment, the allelopathic action of different concentrations of aqueous extracts of eucalyptus leaves on the germination of pitaia seeds was evaluated. This experiment was carried out in DIC with four repetitions of 60 pitaia seeds. These seeds were then subjected to treatments with different dilutions, varying from 0% (control), 12.5%, 25% and 50% of the stock solution. It was observed that as the leaf extract concentrations increased, germination decreased. The concentrations of 0% (control) and 12.5% showed better results. In conclusion, in fact, eucalyptus exerted an allelopathic action on pitaia plants.

Eugenia stipitata occurs along rivers in Western Amazonia and produces berry-type fruits with economic potential. Its large recalcitrant (i.e. desiccation-intolerant) seeds have been proposed as a model to study seed stress response, as no apparent differentiation between the embryonic axis and the fused cotyledons are visible. Here, the longevity of submerged seeds was analysed with a view to understanding adaptive mechanisms to seasonal flooding. Submerged seeds began germinating after 2 months. After 1 year, 87 and 96% total germination was reached when seeds were submerged under a water column of 6 cm (where seedlings could emerge from under the water) and 26 cm (where seedlings could not reach the water surface), respectively. Seedling morphology was altered underwater, with short internodes and rudimentary leaf blades, and when submersion was terminated, seedlings transplanted to nursery conditions recovered a normal phenotype. Furthermore, when seedlings were detached from the seeds, the 'resown' seeds produced a second, normal seedling within 9 months. Concentrations of the antioxidant glutathione, which was measured as a stress marker, increased with submersion time in water. Seeds that had developed roots and shoots underwater had higher concentrations of glutathione disulphide than non-germinated seeds, suggesting that the flooding stress was more intense for seedlings than seeds, although more oxidizing cellular redox environments are also consistent with the conditions required for differentiation. Submergence underwater is recommended for storage of the recalcitrant seeds of E. stipitata for up to 1 year.

The use of biochemical seed viability markers is often compromised by the unknown partitioning of analytes in bulk seed lots consisting of inseparable populations of viable and nonviable seeds. We took advantage of an unusual morphological syndrome found in the recalcitrant, undifferentiated seeds of Eugenia stipitata: one seed can be cut into several parts, each of which can germinate and develop into seedlings. We used four seed parts from one individual seed to analyse seed moisture content (MC), seed viability and the antioxidant glutathione (γ-glutamyl-cysteinyl-glycine; GSH), glutathione disulphide (GSSG) and intermediates of glutathione synthesis and breakdown. Seeds were exposed to different environmental MC to induce various levels of desiccation stress. Upon storage at high seed MC, seed viability was maintained, while GSH concentration increased and the glutathione half-cell reduction potential (E ) was less negative than -215 mV, indicating GSH production and highly reducing conditions. Storage at low seed MC led to loss of GSH, resulting in a shift in E , and seed death. In contrast, the cyst(e)ine half-cell reduction potential (E ) could not distinguish between the viability categories. Previous studies on seed populations revealed that the probability for a seed being alive is 50% at E values between -180 and -160 mV. The single seed approach revealed that the window in which seed viability was lost could be slightly shifted towards more negative values. We discuss the contribution of cellular pH to E and recommend E. stipitata as a recalcitrant seed model to study stress response on a single seed basis.
