Effects of budlein A on human neutrophils and lymphocytes

Abstract: Sesquiterpene lactones (SLs) are the active constituents of a variety of medicinal plants used in traditional medicine for the treatment of inflammatory diseases and other ailments. Objective In this study, we evaluated whether budlein A modulates the activation of innate and adaptive immune cells such as neutrophils and lymphocytes.Material and Methods Our research group has investigated several plant species and several compounds have been isolated, identified, and their medical potential evaluated. Budlein … Show more

“…These inflammatory mediators are detected by vagus nerve and inform the brain about peripheral inflammation (Tracey, 2002; Watkins et al, 1995). IL-6, TNF-α, IL-12, IL-1 alpha and CXCl1 have all previously been shown to play a detrimental role in stroke (Boutin et al, 2001; Losy et al, 2005; Whiteley et al, 2009) and are secreted by monocytes, macrophages and neutrophils (Cai et al, 2016; Chimen et al, 2017; Farahi et al, 2017; Knob et al, 2016; Nowicki et al, 2017; Sica et al, 2012). Interestingly, splenectomy in both sham and stroke animals resulted in a decline in pro-inflammatory cytokines, suggesting a role of spleen in cytokine modulation.…”

“…Moreover, strategies aiming at controlling the initial recruitment of neutrophils are interesting approaches for pain relief in gout arthritis. Previous evidence shows that budlein A inhibits neutrophil recruitment ( Valerio et al, 2007 ; Arakawa et al, 2008 ; Nicolete et al, 2009 ; Knob et al, 2016 ; Zarpelon et al, 2017 ) and activation as observed by a reduction in neutrophil elastase release ( Arakawa et al, 2008 ), an enzyme released in the neutrophil extracellular traps (NETs) ( Fattori et al, 2016 ). In the present work, we demonstrate that budlein A reduced MSU-induced neutrophil recruitment, as observed in the reduced number of LysM-eGFP positive cells in the knee joint, decreased levels of MPO activity, and reduced number neutrophils in the Neubauer chamber.…”

“…Gardner (Brazilian Arnica , Asteraceae) containing the SL goyazensolide (the major metabolite) ameliorated oxonate-induced hyperuricemia in mice, indicating that they could be also used as urate-lowering therapies ( de Albuquerque Ugoline et al, 2017 ). Budlein A is an SL with analgesic ( Valerio et al, 2007 ; Zarpelon et al, 2017 ) and anti-inflammatory properties ( Valerio et al, 2007 ; Arakawa et al, 2008 ; Nicolete et al, 2009 ; Knob et al, 2016 ; Zarpelon et al, 2017 ) related to the inhibition of pro-inflammatory cytokines and neutrophil recruitment. Mechanistically, SLs can inhibit NF-κB activation ( Lyss et al, 1998 ; Siedle et al, 2004 ; Zhao et al, 2015 ; Widen et al, 2017 ; Zarpelon et al, 2017 ) or even direct target inflammasome assembly ( Juliana et al, 2010 ; Mathema et al, 2012 ).…”
