Comparison of primary human gingival fibroblasts from an older and a young donor on the evaluation of cytotoxicity of denture adhesives

Abstract: Denture adhesives (DA) improve the retention and stability of ill-fitting dentures, especially for older adults. These materials should be biocompatible, i.e., they cannot cause undesired biological responses and be non-cytotoxic to oral tissues. However, in vitro testing of DA biocompatibility employing primary cell culture may possibly be affected by other factors, such as the donor age.ObjectiveTo compare the cytotoxicity of three different denture adhesives when assessed in primary gingival fibroblasts fro… Show more

“…The selection of the cell types used in our work was made based on the fact that gingival keratinocytes and fibroblasts are among the most abundant resident cells in the oral mucosa which will have a direct contact with our proposed products. Human gingival fibroblasts exhibit a higher ability to induce scarless wound healing compared to human skin fibroblasts, because of the different release of growth factors [ 87 ].…”

“…The divergence between the results of these studies may be caused by modifications in the experimental parameters measured, such as cell material contact method, cell type, time points, or the preparation of solid specimens or eluates 18 . Several reports point to differences in cytotoxicity that depended on the cell lines and assay techniques used 1,4,18 . This is why human oral cells were used in this study.…”

“…Many patients with dentures use denture adhesives daily because they provide retention and stability, improving both function and comfort 1‐3 . Moreover, the product is easily available at any pharmacy, with no need for a prescription and at an affordable price 4 .…”

“…The gingival fragments were digested with 2 ml type I collagenase solution in PBS (25 U/ml; Sigma Aldrich; Merck KGaA) at 37˚C for 1 h, followed with a 15-min incubation at 37˚C in trypsin-EDTA solution (0.25%; Thermo Fisher Scientific, Inc.). Subsequently, the fragments were incubated in DMEM with 10% FBS (both Sigma Aldrich; Merck KGaA) and 1% penicillin/streptomycin at 37˚C (22). The cultured cells from gingiva were tested by immunocytochemical analysis.…”
