Molecular cloning, characterization and expression analysis of a novel PDRG1 gene from black tiger shrimp (Penaeus monodon)

Abstract: P53 And DNA Damage-Regulated Gene 1 (PDRG1) is a novel gene which plays an important role in chaperone-mediated protein folding. In the present study, the full-length complementary DNA (cDNA) sequence of the PDRG1 gene from Penaeus monodon (PmPDRG1) was cloned by the rapid amplification of cDNA ends (RACE) method. The cDNA of PmPDRG1 spans 1,613 bp, interrupted by only one short intron, and encodes a protein of 136 amino acids with calculated molecular weight of 15.49 kDa. The temporal expression profile of Pm… Show more

“…Although the short type PDRG1 is rare, the alignment results showed that the deduced amino acid sequence QEIVDLDTKRNQNREALRL (30–48 aa) of As -PDRG1 shares high homology with PDRG1 proteins from other species. However, the function difference between the short and long types of PDRG1 is unclear, and further research is needed to determine the significance of this difference [ 20 ]. As an important protein for the cell cycle, As -PDRG1 is ubiquitously expressed in the nucleus, mitochondria, and cytoplasm.…”

“…The primers for real-time PCR (RT- pdrg 1F, RT- pdrg 1F; Table 2 ) were synthesized by TsingKe. The A. sinica β-actin gene (β-actin-F, β-actin-F; Table 2 ) was used as a normalization control for each RNA sample [ 19 , 20 ]. Real-time PCR was performed in triplicate for each sample using SYBR ® Premix Ex Taq™ (Takara).…”

“…In response to pressure signals, cytoplasmic p53 translocated rapidly to mitochondria, where it interacted with multidomain members of the pro-and anti-apoptotic bcl-2 family members to either inhibit or activate them (Vaseva and Moll, 2009). p53 and DNA damage regulated gene (PDRG) are the genes which are associated with apoptosis and cell cycle regulation (Luo et al, 2003;Jiang et al, 2011;Wang et al, 2014;Zhao et al, 2017). It is known that PDRG appears as a specific marker in DNA damage and be selectively regulated by agents that induced genotoxic stress in vertebrates (Luo et al, 2003;Jiang et al, 2011).…”
