Assesment of the Genotoxic Effect of the Diazinon on Root Cells of Allium cepa (L.)

Os agrotóxicos são produtos químicos sintéticos amplamente utilizados no âmbito agrícola para controlar pragas e doenças. O modelo de agricultura predominante no Brasil baseia-se no uso demasiado desses produtos e atualmente o país ocupa o primeiro lugar no ranking mundial de consumo. Apesar de possibilitar o crescimento da produção agrícola, a utilização excessiva de agrotóxicos causa consequências à saúde humana e ao meio ambiente. O objetivo deste estudo foi avaliar os possíveis efeitos tóxicos, citotóxicos e genotóxicos do inseticida ciromazina utilizando o bioensaio com Allium cepa. Os bulbos saudáveis de cebola foram expostos a diferentes dosagens do inseticida, além dos controles positivo e negativo por um período de cinco dias. Após o qual, foi realizada a análise macroscópica para a avaliação da toxicidade, sendo contabilizado o número de raízes germinadas e o comprimento das três maiores raízes de cada bulbo. Para a avaliação da citotoxicidade e genotoxicidade, foram confeccionadas lâminas para análise das células em diferentes fases do ciclo celular. O inseticida testado foi tóxico para A. cepa, inibindo a germinação e o crescimento radicular em diferentes concentrações averiguadas. Além disso, foram observados efeitos citotóxicos nas duas maiores concentrações analisadas. O composto foi considerado genotóxico, pois induziu aberrações cromossômicas e micronúcleos nas células, especialmente nas maiores dosagens. O estudo contribuiu com informações relevantes acerca da toxicidade e genotoxicidade do inseticida ciromazina em A. cepa, mas também indica a necessidade de pesquisas adicionais em outros organismos teste.

Öz: Yaygın olarak kullanılan bir pestisit olan diazinon hedef olmayan canlılar üzerinde toksik etkilere sahiptir. Bu çalışma, diazinonun meristem hücrelerinde teşvik ettiği toksisiteye karşı zencefilin koruyucu ve iyileştirici etkisini fizyolojik, genotoksik ve biyokimyasal parametrelerle araştırmayı hedeflemiştir. Ayrıca kök meristem hücrelerinde meydana gelen hasarlar incelenmiştir. Bu amaçla A. cepa soğanları 6 gruba ayrıldı (kontrol, 240 mg/L zencefil, 480 mg/L zencefil, 500 mg/L diazinon, 500 mg/L diazinon + 240 mg/L zencefil ve 500 mg/L diazinon + 480 mg/L zencefil). Diazinon süperoksit dismutaz aktivitesi, katalaz aktivitesi, malondialdehit seviyesi, mikronükleus sıklığı, kromozomal anormallik yoğunluğu ve meristematik hücre hasarlarını arttırmış; ancak mitotik indeksi, çimlenme yüzdesi, kök uzunluğu, ağırlık artışı azaltmıştır. Zencefil, diazinon ile birlikte uygulandığında doza bağlı olarak tüm bu toksisite göstergelerini düşürmüştür. Bu çalışma, A. cepa modeli kullanılarak diazinonun hedef olmayan organizmalar üzerindeki toksik etkilerine karşı zencefilin koruyucu bir rolünü açıkça göstermiştir.

The comet assay is a sensitive method for the evaluation of DNA damages and DNA repair capacity at single‐cell level. Allium cepa is a well‐established plant model for toxicological studies. The aim of this scoping review was to investigate the recent application of the comet assay in Allium cepa root cells to assess the genotoxicity. To explore the literature a search was performed selecting articles published between January 2015 and February 2023 from Web of Science, PubMed, and Scopus databases using the combined search terms “Comet assay” and “Allium cepa”. All the original articles that applied the comet assay to Allium cepa root cells were included. Of the 334 records initially found, 79 articles were identified as meeting the inclusion criteria. Some studies reported results for two or more toxicants. In these cases, the data for each toxicant were treated separately. Thus, the number of analyzed toxicants (such as chemicals, new materials, and environmental matrices) was higher than the number of selected papers and reached 90. The current use of the Allium‐comet assay seems to be directed towards two types of approach: the direct study of the genotoxicity of compounds, mainly biocides (20% of analyzed compounds) and nano‐ and microparticles (17%), and assessing a treatment's ability to reduce or eliminate genotoxicity of known genotoxicants (19%). Although the genotoxicity identified by the Allium‐comet assay is only one piece of a larger puzzle, this method could be considered a useful tool for screening the genotoxic potential of compounds released into the environment.
