The anesthetic agent sevoflurane attenuates pulmonary acute lung injury by modulating apoptotic pathways

Abstract: The objective of this study was to evaluate lung protection by the volatile anesthetic sevoflurane (SEVO), which inhibits apoptosis. Male Sprague-Dawley rats (250–280 g; n=18) were randomly divided into three groups. The LPS group received 5 mg/kg endotoxin (lipopolysaccharide), which induced acute lung injury (ALI). The control (CTRL) group received normal saline and the SEVO group received sevoflurane (2.5%) for 30 min after ALI was induced by 5 mg/kg LPS. Samples were collected for analysis 12 h after LPS. … Show more

“…The caspase-3 activity is an important marker of cellular apoptosis. Increased activation of caspase-3 has been reported in the animal models of ALI [28,29]. In the current study, enhanced caspase-3 activity, elevated production of IL-1b and IL-6, together with the increased apoptosis ratio was observed following CLP operation.…”

“…The balance between pro-apoptotic and anti-apoptotic Bcl-2 family members determines the mitochondrial response to apoptotic stimuli [ 14 ]. The imbalance between pro-apoptotic and anti-apoptotic mediators causes apoptosis and increases susceptibility to lung injury [ 15 ].…”

“…Finally, we examined the epithelial and endothelial cell apoptosis, since it has been shown that increased apoptosis reflects the degree of tissue damage in lungs from ALI patients (26,27). To determine whether ACGs diminished apoptosis (and subsequent tissue damage) in ALI mice, we employed TUNEL staining to quantify the number of apoptotic cells in lung tissues (Fig.…”
