Additive effects of CuSO4 and aromatic compounds on laccase production by Pleurotus sajor-caju PS-2001 using sucrose as a carbon source

Abstract: -Laccase enzymes are now commercially available, and a laccase/mediator combination is currently marketed for indigo dye bleaching in textile manufacturing; replacing traditional chemical-based processes with enzymatic technology reduces the need for effluent treatment. However, an inexpensive source of these enzymes will be needed to enable wider application of this technology. In the present work, the main objective was to increase laccase production by the mushroom Pleurotus sajor-caju strain PS-2001 grown … Show more

“…During the experiments, three different conditions of oxygen concentration (DO) were tested (uncontrolled, 30% and 80% of saturation in the medium), using basic medium constituted of glucose, pure casein and mineral solution added of CuSO4 and benzoic acid as enzymatic inducers at pH controlled between 4.5 and 7.0 [28,29]. Figure 1 shows laccases activities, substrate and biomass concentrations during the 168 hours of the experiment.…”

“…For the production of laccase-containing enzyme broth, the following cultivation medium was used (per litre): glucose, 5.0 g; pure casein (Synth ® ), 1.5 g; CuSO4, 100 mg; benzoic acid, 100 mg; mineral solution, 100 mL [32]. The cultivations were carried out in a stirred-tank bioreactor B. Braun Biotech model Biostat ® B with 4.0 L of working volume.…”

“…Due to their broad industrial and environmental uses, these enzymes have gained increasing attention. Laccase production can be stimulated by various inductive substances, including metal ions, aromatic or phenolic compounds, alcohol, and detergents (Collins and Dobson, 1997;Birhanli and Yesilada, 2006;Tychanowicz et al, 2006;Bettin et al, 2014). Furthermore, inducer concentration, fermentation types, growth media, and incubation conditions have an important effect on laccase production and they are specific to species and strains (Boran and Yesilada, 2011).…”
