Actin expression in trypanosomatids (Euglenozoa: Kinetoplastea)

Abstract: Heteroxenic and monoxenic trypanosomatids were screened for the presence of actin using a mouse polyclonal antibody produced against the entire sequence of the Trypanosoma cruzi actin gene, encoding a 41.9 kDa protein. Western blot analysis showed that this antibody reacted with a polypeptide of approximately 42 kDa in the whole-cell lysates of parasites targeting mammals (T. cruzi, Trypanosoma brucei and Leishmania major), insects (Angomonas deanei, Crithidia fasciculata, Herpetomonas samuelpessoai and Strigo… Show more

“…In trypomastigotes, TcAct1 was uniformly distributed with a low level of staining ( Cevallos et al, 2011 ), and in the cell-derived amastigotes, a heterogeneous TcAct1 localization with sometimes no apparent expression was observed ( Cevallos et al, 2011 ). Similar protein expression levels and intracellular TcAct1 distributions in epimastigotes, amastigotes and metacyclic trypomastigotes were also observed by using mouse polyclonal anti-recombinant TcAct1 antibodies ( Souza et al, 2013 ). In addition to TcAct1, TcAct2 has also been characterized.…”

“…Following protein transfer, the membranes were blocked with 5% non-fat milk/0.05% Tween-20 in PBS and incubated for 1 h with blocking buffer containing either the antiserum raised against the TcCLC protein (1:200 dilution) or the monoclonal antibody against TcCHC. A polyclonal anti-TcActin antibody [ 37 ] was used as a loading control. After 3 washes with 0.05% Tween-20/PBS, the membranes were incubated for 1 h with alkaline phosphatase-conjugated rabbit anti-mouse IgG (1:10,000 dilution), washed three times with 0.05% Tween-20/PBS and bands then visualized using BCIP-NBT (Promega, USA) solution.…”

“…The membrane was washed three times in 0.05% Tween-20/PBS and then incubated for 1 h with AP-conjugated rabbit anti-mouse IgG (diluted 1 : 10,000). A polyclonal antirecombinant actin (TcActin; diluted at 1 : 200) mouse serum [ 23 ] was used for normalization. The membrane was then washed three times with 0.05% Tween-20/PBS and the reactive bands were visualized with BCIP-NBT solution, as described by the manufacturer.…”
