Context Callistemon citrinus Skeels (Myrtaceae) exhibits many biological activities. Objective This study analyzes for the first time, the toxicity, obesogenic, and antioxidant effects of C. citrinus in rats fed with a high fat-fructose diet (HFFD). Materials and methods Four studies using male Wistar rats were conducted: (a) 7 groups ( n = 3): control (corn oil) and ethanol extract of C. citrinus leaf (single oral dose at 100–4000 mg/kg) for acute toxicity; (b) 2 groups ( n = 8): control (corn oil) and C. citrinus (1000 mg/kg/day) for 28 days for subacute toxicity; (c) 3 groups ( n = 4) with single oral dose of lipid emulsion: control (lipid emulsion), C. citrinus and orlistat (250 and 50 mg/kg, respectively) for lipid absorption; (d) 4 groups ( n = 6): control (normal diet) and 3 groups fed with HFFD: HFFD only, C. citrinus and simvastatin (oral dose 250 and 3 mg/kg, respectively) for 13 weeks. Antioxidant enzymes and biomarkers were evaluated and inhibition of pancreatic lipase was determined in vitro . Results Toxicological studies of C. citrinus showed no differences in biochemical parameters and lethal dose (LD 50 ) was higher than 4000 mg/kg. C. citrinus inhibited pancreatic lipase activity, with IC 50 of 392.00 µg/mL, and decreased lipid absorption by 70%. Additionally, it reduced the body weight 22%, restored the activities of antioxidant enzymes, and reduced the biomarkers of oxidative stress. Conclusions Callistemon citrinus showed an effect against oxidative stress by reducing biomarkers and induced antioxidant system, without toxic effects.

Background Pesticide toxicity is considered to be one of the significant reason for increased incidence of cancer. Plants are treasure troves of active phytochemical compounds which are used as herbal medicine as well as nutraceuticals. Objective To evaluate the genoprotective potential of Orthosiphon thymiflorus (Roth) Sleesen, (Lamiaceae) against the toxicity induced by malathion by a battery of four in vivo assays in Swiss albino mice. Materials and methods Micronucleus assay was performed for analyzing the micronuclei induction and ratio of polychromatic and normochromatic erythrocytes (PCE/NCE). Anticlastogenic and mito depressive effect of the methanol and hexane extracts of O.thymiflorus were evaluated by chromosome aberration assay. Alkali comet assay was performed to assess double strand DNA repair. DNA damage sensing ability of the bone marrow cells were assessed by γ-H2AX foci formation. Phytochemical screening of hexane and methanol extract was done by GC–MS analysis. Result O. thymiflorus extracts showed a dose dependant protective effect in all assays. It significantly decreased the frequency of micronuclei and improved PCE/NCE value in post treated groups of animals. Malathion induced clastogenic aberrations were effectively attenuated by methanol and hexane extracts. DNA comet assay showed that malathion induced damage can be protected by O. thymiflorus extracts. Multiple foci formation in γ-H2AX assay confirmed the activation of DNA repair proteins in post treated animals. Conclusion Genoprotective effect of O. thymiflorus against malathion induced toxicity was confirmed. This study would be helpful to initiate more research including clinical using O. thymiflorus extract against pesticide induced toxicity.

Ornamental plants often gain relevance not only for their decorative use, but also as a source of phytochemicals with interesting healing properties. Herein, spontaneous Centranthus ruber (L.) DC. and Tropaeolum majus L., mainly used as ornamental species but also traditionally consumed and used in popular medicine, were investigated. The aerial parts were extracted with methanol trough maceration, and resultant crude extracts were partitioned using solvents with increasing polarity. As previous studies mostly dealt with the phenolic content of these species, the phytochemical investigation mainly focused on nonpolar constituents, detected with GC-MS. The total phenolic and flavonoid content was also verified, and HPTLC analyses were performed. In order to explore the potential antiarthritic and anti-obesity properties, extracts and their fractions were evaluated for their anti-denaturation effects, with the use of the BSA assay, and for their ability to inhibit pancreatic lipase. The antioxidant properties and the inhibitory activity on the NO production were verified, as well. Almost all the extracts and fractions demonstrated good inhibitory effects on NO production. The n-hexane and dichloromethane fractions from T. majus, as well as the n-hexane fraction from C. ruber, were effective in protecting the protein from heat-induced denaturation (IC50 = 154.0 ± 1.9, 270.8 ± 2.3 and 450.1 ± 15.5 μg/mL, respectively). The dichloromethane fractions from both raw extracts were also effective in inhibiting pancreatic lipase, with IC50 values equal to 2.23 ± 0.02 mg/mL (for C. ruber sample), and 2.05 ± 0.02 mg/mL (T. majus). Obtained results support the traditional use of these species for their beneficial health properties and suggest that investigated plant species could be potential sources of novel antiarthritic and anti-obesity agents.
