cDNA sequence analysis of a 29-kDa cysteine-rich surface antigen of pathogenic Entamoeba histolytica.

Abstract: A Agtll cDNA library was constructed from poly(U)-Sepharose-selected Entamoeba histolyica trophozoite RNA in order to clone and identify surface antigens. The library was screened with rabbit polyclonal anti-E. histolytica serum. A 700-base-pair cDNA insert was isolated and the nucleotide sequence was determined. The deduced amino acid sequence of the cDNA revealed a cysteine-rich protein. DNA hybridizations showed that the gene was specific to E. histolytica since the cDNA probe reacted with DNA from four axe… Show more

“…Construction of an E. histolytica cDNA Agtll library from strain H-302:NIH has been described elsewhere (9). Recombinant bacteriophage was grown on Petri plates and overlaid with a nitrocellulose filter that had been soaked in isopropyl f-Dthiogalactopyranoside as described in the ProtoBlot protocol (Promega) and in ref.…”

“…Two other ORFs are each similar to several proteins in the Swiss-Prot and GenPept data bases, but still defy identification. ORF 199 is similar to a mouse erythroleu kemia cell line-specific protein (MER5) (Yamamoto et al, 1989), a Helicobacterpyloriantigen (OToole et al, 1991), an Entamoeba histolytica surface antigen (Torian et al, 1990), and Salmonella typhimurium alkyl hydroperoxide reductase (Tartaglia et al, 1990). ORF 587 is similar to a group of eukaryotic proteins, Paslp, Secl8p, NSF, Cdc48p, and VCP, that have a conserved domain involved in ATP hydrolysis (Erdmann et al, 1991).…”

“…Mice were given two booster injections with 100 p.g of OmpF trimers at 3-week intervals. The cell fusion and cloning by limiting dilution were performed as described by Torian et al (41,42). Screening was performed by ELISA with purified OmpF trimers and monomers at 1 p.g per well.…”
