Heterologous expression of a rice metallothionein isoform ( OsMTI-1b ) in Saccharomyces cerevisiae enhances cadmium, hydrogen peroxide and ethanol tolerance

Abstract: Metallothioneins are a superfamily of low-molecular-weight, cysteine (Cys)-rich proteins that are believed to play important roles in protection against metal toxicity and oxidative stress. The main purpose of this study was to investigate the effect of heterologous expression of a rice metallothionein isoform (OsMTI-1b) on the tolerance of Saccharomyces cerevisiae to Cd2+, H2O2 and ethanol stress. The gene encoding OsMTI-1b was cloned into p426GPD as a yeast expression vector. The new construct was transforme… Show more

“…Abiotic stresses like HMs induce excessive accumulation of ROS in plants, and cause damages to the cellular macromolecules such as proteins, leading to metabolic and physiological disorders in cells or even cell death (Hasan et al, 2016 ). Interestingly, MTs have been proposed as an alternative tool by which plants protect themselves from stress-induced oxidative damage (Figure 1 ; Hassinen et al, 2011 ; Ansarypour and Shahpiri, 2017 ). Although many reports have indicated the roles of MTs in abiotic stress tolerance as ROS scavengers, the mechanisms through which MTs mediate ROS homeostasis remain unclear (Hassinen et al, 2011 ).…”

“…In other studies, enhanced tolerance towards ROS or compounds that induce ROS production in non-hypersensitive to oxidative stress yeast cells expressing pMTs was observed (Ansarypour & Shahpiri, 2017;Nishiuchi, Liu, & Takano, 2007;Samardžić et al, 2010;Xue et al, 2009). However, both our and Hegelund et al (2012) results indicated that in yeast mutant strains that are oversensitive to oxidative stress, yap-1Δ and skn7Δ, respectively, expression of pMTs cannot complement or even intensifies the loss of H 2 O 2 tolerance.…”
