Centenarians, or individuals who have lived more than a century, represent the ultimate model of successful longevity associated with decreased susceptibility to ageing-associated illness and chronic inflammation. The gut microbiota is considered to be a critical determinant of human health and longevity. Here we show that centenarians (average 107 yo) have a distinct gut microbiome enriched in microbes capable of generating unique secondary bile acids, including iso-, 3-oxo-, and isoallo-lithocholic acid (LCA), as compared to elderly (85-89 yo) and young (21-55 yo) controls. Among these bile acids, the biosynthetic pathway for isoalloLCA had not been described previously. By screening 68 bacterial isolates from a centenarian’s faecal microbiota, we identified Parabacteroides merdae and Odoribacteraceae strains as effective producers of isoalloLCA. Furthermore, we generated and tested mutant strains of P. merdae to show that the enzymes 5α-reductase (5AR) and 3β-hydroxysteroid dehydrogenase (3βHSDH) were responsible for isoalloLCA production. This secondary bile acid derivative exerted the most potent antimicrobial effects among the tested bile acid compounds against gram-positive (but not gram-negative) multidrug-resistant pathogens, including Clostridioides difficile and vancomycin-resistant Enterococcus faecium. These findings suggest that specific bile acid metabolism may be involved in reducing the risk of pathobiont infection, thereby potentially contributing to longevity.

The application of a bronchoconstrictor, usually Methacholine (MCh), in respiratory mechanics studies is usually accompanied by the assessment of respiratory mechanics in a dose–response curve. The MCh used in the dose–response curve can be inhaled (i.h.) and intravenous (i.v.) and there are studies comparing i.v. bolus and i.h. MCh in both mice and rats. However, MCh i.v. can be injected at short time interval (bolus) or in continuous infusion. This comparison is relevant since the way MCh is applied influences the mathematical model. We chose an aging process scenario to compare both protocols. This study aims to compare respiratory mechanics of 3-, 6-, and 10-month SAMR1 mice and how both administration methods (continuous infusion and bolus) impact respiratory mechanics evaluation. Both protocols were capable of assessing the difference among ages and doses in: peak or plateau; and area under the curve analysis. The respiratory mechanics parameters were Rn, G, and H (two-way analysis of variance: groups and doses with a P < 0.05 for all). Also, the infusion protocol presented a higher sensitivity to dose increment. In conclusion, both protocols were able to discriminate intragroup and intergroup differences. In the bolus protocol, the highest value of each curve dose may not correspond to the highest real value, and the loss of this point may be a problematic factor in the sample size. These factors are not present in the infusion protocol. Additionally, at this lineage and age screening, the infusion protocol appeared to be more sensitive to differences among ages when compared to the bolus protocol. Impact statement Respiratory mechanics studies are associated with fundamental research and translational studies; the present work thus investigates this particular matter. Our current research describes differences and similarities between two different ways of administrating a very prevalent bronchoconstrictor (methacholine) in an aging process scenario. The core issue of our work is related with troubles we find with the bolus protocol and the application of the mathematical model used to assess the respiratory mechanics. Our findings reveal the continuous infusion as an alternative to these problems and we hope to provide the proper foundations to a more reliable assessment in the respiratory field.

Background: No papers have examined the relationship between socio-demographic characteristics and cognitive performance in oldest old subjects (i.e, > = 80 years old) asking for driving license renewal. We hypothesize that, even in this highly functioning population, age, sex, and education influence cognitive performance, expressed as total or single domain (raw) test scores. This research question allows to describe, identify, and preserve independence of subjects still able to drive safely. Methods: We examined cross-sectionally a cohort of > = 80 years old subjects (at enrollment) asking for driving license renewal in the Milan area, Italy, 2011-2017. The analysis was restricted to 3378 first and 863 second visits where individual's cognitive performance was evaluated. According to the study protocol, the Mini Mental State Examination (MMSE) test was administered at the first visit for driving license renewal and the Montreal Cognitive Assessment (MoCA) test at the second visit, following an additional renewal request. Ordinary least squares regression models were fitted at either time points. In each model, we included age, sex, and education as independent variables, whereas the dependent variable was total or single domain score for either test. In total, we fitted 15 regression models to assess our research hypothesis. Results: The median subject in our sample reached the maximum scores on domains targeting operational and tactical abilities implied in safe driving, but had sub-optimal scores in the long-term memory domain included among the strategic abilities. In multiple models, being > = 87 (versus 80-< 86 years old) significantly decreased the mean total and memory scores of MMSE, but not those of the MoCA. Females (versus males) had significantly higher mean total and long-term memory scores of either tests, but not other domains. Mean total and single domain scores increased for increasing education levels for either tests, with increments for high school graduates being~2 of those with (at most) a junior high school diploma.
