1-Phosphatidylinositol 3-kinase activity is required for insulin-stimulated glucose transport but not for RAS activation in CHO cells.

Abstract: Insulin stimulation drives the formation of a complex between tyrosine-phosphorylated insulin receptor substrate 1 (IRS-1) and 1-phosphatidylinositol 3-kinase (PI 3-kinase; ATP:l-phosphatidyl-lD-myo-inositol 3-phosphotransferase, EC 2.7.1.137), a heterodimer coIng of regulatory 85-kDa (p85) and catalytic 110-kDa (p11O) subunits. appears to be the subunit that links PI 3-kinase activity in pilO to the tyrosine-phosphorylated proteins. The insulin receptor belongs to the family of structurally related transmembra… Show more

“…PAK1 (P191G,R192A) tagged with myc epitope at the C-terminus was generated and cloned into the pCS2+ vector. Wild-type and mutated forms (Dp85, p85(III)) of bovine p85a, and p110 subunit of PI3-kinase with myristoylation signal (myr p110) were provided by W Ogawa (Yonezawa et al, 1992;Hara et al, 1994). Plasmid encoding p130 cas was provided from H Hirai (Nakamoto et al, 1996), and chicken p125 FAK from JT Parsons.…”
